Liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI MS/MS) combines the separation capabilities of liquid chromatography with ionization power of electrospray ionizer and mass analysis capability and specificity of tandem mass spectroscopy. Electrospray ionization is very useful in producing ions from macromolecules as it overcomes their tendency to fragment upon ionization, and it can also produce multiply charged ions which extend the mass analysis range of analyzer to accommodate masses of kilo-Dalton to mega-Dalton. Liquid chromatography helps to separate components in sample according to their size and relative affinity to mobile and stationary phase. Upon elution, the components are ionized by electrospray ionizer and enter the two mass spectrometers placed in series. They analyze the components and produce a mass spectrum showing their mass-to-charge ratio and relative abundance which helps in identifying and quantifying components in sample mixture.
