In high-performance liquid chromatography mass spectrometry (HPLC-MS), the separation capability of HPLC is coupled with mass analysis capability of mass spectrometry via a suitable interface such as electrospray ionization. Sample is introduced in the system through an injection port. A high pressure flow of solvent (mobile phase) makes the sample pass through a column packed with coated beads (stationary phase). As components in the sample are separated based on their relative affinity to stationary phase and mobile phase, they are introduced into the mass spectrometer, where they are ionized to produce charged molecules. These move through a vacuum chamber as they are accelerated with electric and magnetic fields, and arrive at the detector according to their mass-to-charge ratios. A mass spectrum is produced which is used for identification and quantification of the compounds in sample. Thermally labile compounds as well as those with very high polarity or high molecular mass are detected and analyzed using this setup.
