Gel electrophoresis liquid chromatography-mass spectrometry (GeLC-MS) involves separation of proteins in sample by gel electrophoresis (SDS-PAGE) followed by the excision of entire gel lanes and in-gel digestion with trypsin. The resulting peptides are loaded in a liquid chromatography system coupled to a mass spectrometer. Electrophoresis separates proteins by size and liquid chromatography separates the resultant peptides by their size and relative affinity to the mobile and stationary phase. They are detected and characterized in mass spectrometer by their relative abundance and mass-to-charge ratio. The results produced by GeLC-MS system are compared to a database of biological samples for analysis and helps in identification and quantification sample constituents.
