Embryonic stem cells require special conditions in order to be maintained in their pluripotent state. Early culture methods relied upon plating ES cells upon a feeder layer, usually of mouse embryonic fibroblasts. Later, it was determined that these feeder cells provided a factor, LIF, that blocked differentiation of ES cells. This has led to the development of feeder-free ES culture methods that can sometimes be used instead of a feeder cell layer.
