Bacterial artificial chromosome (BAC) DNA is isolated with BAC DNA extraction. A BAC is a circular DNA construct based on the F-plasmid (functional fertility plasmid) and contains the inserted gene of interest (150-350 kbp) as well as partition genes that code for even plasmid distribution after bacterial cell division. The BACs are then taken up by comparable bacterial cells (usually E. coli) and amplified through bacterial growth. BACs are extracted using a modified alkaline-lysis protocol and can then be sequenced. Because BACs accommodate large sequences that do not undergo rearrangement, they are an especially stable cloning vector ideal for sequencing entire organisms' genomes and for modeling genetic diseases.
