zPREDICTA offers physiologically relevant cell culture models that recapitulate the major stages of human cancers. Utilizing the 3D tissue culture systems and spheroid assays, we help our clients study tumor formation, invasion, and metastasis – from studies of basic biology to target discovery and complex preclinical screens of investigational compounds and drug combinations. Our 3D cultures are customized to the tissue and cancer of interest and provide researchers access to enriched cell populations representing the primary tumor, as well as invasive, metastatic, and cancer stem cells.

Our models incorporate tissue microenvironment of the primary tumor and the distant metastatic site recapitulating the elements of environment-mediated drug-resistance in the context of:

• tissue-specific extracellular matrix (ECM)
• stromal-derived soluble factors
• tumor-tumor interactions
• tumor-stroma interactions

Our culture systems have been validated both in vitro and in vivo and were tested on multiple cancer cell lines and primary tissues representing multiple types of solid tumors and hematologic malignancies. Our models mimic in vivo behavior of tumors and exhibit high correlation with clinical response.

zPREDICTA has developed organ-specific, physiologically-relevant 3D cell culture models providing a 1-to-1 reconstruction of human tissues. This is the only available 3D culture approach that comprehensively accounts for the major components of the cellular and extracellular tissue microenvironment preserving the critical interactions between a tumor and its surroundings. Clinical study data demonstrate the high correlation between drug efficacy measured in our platform and the clinical response. The ability of the zPREDICTA platform to reliably predict clinical outcomes offers superior compound attrition management, empowering researchers to exclude ineffective compounds while selecting agents with a high probability of success in the clinic. Our approach is compatible with any drug class, multiple tissue/cell types and a wide variety of readouts. Our customers are using these models for efficacy screening of anticancer compounds, including immuno-oncology agents, evaluation of mechanisms of drug resistance, the rescue of failed drug candidates, assessment of off-target toxicity, and a multitude other applications.

PRIMARY CELL MODELS:
• r-Bone-MM (reconstructed bone marrow for 3D culture of primary multiple myeloma cells)
• r-Bone-AML (reconstructed bone marrow for 3D culture of primary AML cells)

CELL LINE & CO-CULTURE MODELS:
• Epithelial tumor cell lines cultured in organ-specific 3D environment (i.e. ECM formulated to mimic lung, breast, prostate, colon, gastric, ovarian, and melanoma tumor microenvironment)
• Epithelial tumor cell lines co-cultured with primary cancer-associated fibroblasts (CAFs), non-malignant fibroblasts, mesenchymal stem cells (MSCs) and/or immune components

In Vitro/Ex Vivo Drug Efficacy Testing

Our 3D cell culture platform is designed to evaluate investigational anticancer therapeutics and drug combinations within the native microenvironment of the human tissues. Our pipeline allows testing new agents under the physiological conditions of environment-mediated drug resistance (EM-DR). We have the capacity to evaluate novel therapeutics on cells of the primary tumor, as well as invasive, metastatic, and cancer stem cell populations.

Utilizing the 3D cell culture platform we can evaluate a number of parameters in the context of environment-mediated drug resistance (EM-DR):
• Identify patient population(s) responsive to the drug of interest
• Screen lead compounds/drug candidates to: 1) establish under various conditions of EM-DR and 2) ascertain the source(s) of resistance, i.e. identify the components of tissue microenvironment that may impede drug function
• Identify drug combinations that can overcome EM-DR
• Evaluate off-target effects and gauge potential toxicity (test cytotoxic effects of investigational compounds on non-malignant cell populations (epithelial, stromal, and hematopoietic))
• Evaluate the capacity of the investigational compounds to target invasive, metastatic and cancer stem cell populations
• Functional fractionation of heterogeneous tumors for target and biomarker discovery
• Determination of growth capacity of various cell populations

ASSAYS:
• Morphology analysis
• Cell proliferation (MTS assay, Ki67 staining, etc.)
• Cell migration (migration through ECM, chemoattraction, etc.)
• Cell viability (MTS assay, annexin V staining, Live/Dead analysis, etc.)
