Developing new drugs costs €500-1000bn and takes 10-15 years. The preclinical phase is critical and expensive (30% total investment). Our solution for cutting time and budget is the use of zebrafish, an animal model that allows to obtain high quality data in a fast and inexpensive manner.

ZeClinics initiates every molecule safety study by testing its Acute Toxicity. This assay allows to assess its possible lethality and teratogenicity and provides values of LC50, EC50 and NOEC.
Assay (under OECD Nº236 guidelines): Incubation of zebrafish embryos in 5 different logarithmic concentrations of the molecule of interest.
Method: Determination of the following parameters at different time-points (24, 48 and 96hpf) of development: EC50, LC50, NOEC (No observed effect concentration), LOEC (Lowest observed effect concentration). And the main parameters of embryopathy/teratogenic: Number of coagulated eggs. Formation of somite. Tail detachment. Presence of heart-beat. Body deformity. Somite morphology. Tail detachment. Aim: Assessment of Cardiotoxicity caused by NOEC of new drugs in zebrafish larvae.

Model validation: Comparison studies of human and zebrafish electrocardiograms show similar ECG patterns and similar impact in cardiac rhythm. Comparison analysis between zebrafish larvae, the cellular human ether-a-go-go-related gene (hERG) and in vivo dog models revealed a strong correlation between zebrafish larvae and hERG model, indicating zebrafish as a strong alternative model for drug cardiotoxicity assessment.

Assay: Incubation of 10-20 Tg(CMLC2::GFP) zebrafish larvae expressing GFP in the heart with the NOEC concentration of the molecule of interest at 72 hpf. At end point every larvae heart is imaged during 1 min and analyzed with ZeCardio system (ZeClinics developed cardio software). Negative control: DMSO. Positive control: Haloperidol. Output: Videos are analyzed for the presence of the following dysfunctions:
•Heart beat frequency (Ventricle and Atrium).
•QT interval.

We propose a number of toxicology assays to test, in a high-throughput manner, the safety of novel drugs. ZeClinics will initiate every molecule safety study by testing its Acute Toxicology. This assay will allow to assess its possible lethality and teratogenicity. On the other hand, by testing a range of 5 logarithmic concentrations, it will provide values of LC50, EC50 and NOEC. Embryos from these treatments can be used optionally to test the ADME of any given molecule. To warrant the safety of any given molecule, and once the NOEC value is established, ZeClinics can proceed to test the putative toxic effects on specific tissues known to be affected by novel drugs: Liver (Hepatotoxicity), Heart (Cardiotoxicity) and CNS (Neurotoxicity).

We are developing the ZeOncoTest, an R&D project aimed at perfecting a cheap, fast and reliable personalized oncological test that will determine the best possible treatment for every individual cancer patient.

The ZeOncoTest uses zebrafish embryos as xeno-hosts for human cancer cells in order to assess the capability of those cells to grow and metastasize under different chemical conditions in an in vivo environment. The product is based on our own research and a broad range of scientific precedents. The ZeOncoTest idea is to transplant primary tumor cells, coming from a cancer patient, into zebrafish embryos. Then, we grow these transplanted embryos under the whole range of possible treatments for that cancer type. In only a few days, and after careful image and statistical analysis, the ZeOncoTest will provide enough information to help the medical team to decide, with a greater chance of success, which of the available treatments is the most suitable to prevent the growth and metastasis of this specific patient cancer.

As stated before, finding the correct treatment from the beginning of the diagnosis will increase enormously the rate of survival and decrease the time of recovery, with its associated suffering. But, also the resources spent during the healing of a cancer patient. In parallel, the ZeOncoTest can be used by pharmaceutical companies, including ZeClinics, as a tool for drug discovery in oncology field. Moreover it can be applied to late phases of drug development to identify the potential sub-group of a given tumor type and accelerate the process of acceptance of new anti-tumoral compounds.

Aim: Assessment of neurotoxicity caused by NOEC of new drugs in zebrafish larvae.
Model validation: zebrafish behaviours include feeding, seizures, involuntary movement, sleeping, and addiction. Therefore, zebrafish is susceptible to both assessing the toxicity of drugs affecting those behaviours or finding novel neuroprotectors.

Assay: Incubation of 20 dechorionated larvae of Tgisl1: GFP with the NOEC concentration of the molecule of interest from 72 hpf to 96 hpf. Locomotor activity is measured through DanioVision™ (Noldus IT). After observation, larvae are fixed and neuronal clusters are observed and analyzed through confocal microscopy and imaging software. Negative control: DMSO. Positive control: MPTP. Output: Determination of different parameters: Basal locomotor activity.
