Purpose
The Vironomics core is a small core facility tailoring our resources to fit your individual project. The Vironomics Core is designed to facilitate research by performing large-scale Real Time PCR reactions and next generation sequencing (NGS) using Ion Torrent S5 and companion Ion Torrent Chef [sequencing on hold till Jan 2019 for retooling]. A Tecan Freedom Evo© is used to set up reactions in a 384-well plate, which is then run by the Roche LightCycler 480 II©. This is a fast and effective way to amplify samples with RT-qPCR.

About Us

The Vironomics Core is led by our Principal Investigator, Dr. Dirk Dittmer, and includes several research staff. Our facilities are located at the University of North Carolina. The main research focus of Dr. Dittmer's lab is understanding viral tumorigenesis, especially that associated with Kaposi's Sarcoma Herpesvirus (KSHV).

NOTE: We are not approved internally for separate rates for the Ion Torrent S5 and Ion Torrent PGM; however, the reagent costs are similar so we will use this rate regardless of sequencing technology.

NGS Data Analysis
Next generation sequencing data analysis
Starting at $106.00 per hour

The rate is per hour. The core will provide data analysis for users, who do not wish to do it themselves. As with all core technologies, data analysis is not straightforward.

This rate also will be applied to projects that require excessive research or R&D to meet the project needs.

Viral Load Determination (clinical)- $182/Sample

This rate will determine virus loads, i.e. how much virus is in a supplied sample. This rate covers sample genomic isolation, cDNA preparation if needed, and analysis of this sample with a single primer and control for sample integrity.. The reporting and regulatory requirements are much more stringent for clinical material. Clinical samples require a heavy investment to conform with federal standards that apply to bio-safety, GLP, HIPPA and GCLP regulations. Note that the core does not accept patient samples, or any samples that can be linked to individual persons. All samples must be de-identified. Research only, information cannot be used to treat or diagnose patients.


Viral load - batch of 8 (experimental) - $215/Run

The rate is per sample (batch of 8) per real-time PCR primer. The core does not accept single samples for this rate. This rate will determine virus loads, i.e. how much virus is in a supplied sample. The Core will provide a central service for quantitative single viral load determination (using real-time QPCR instrument Roche LightCycler 480) of experimental samples, this includes running quality control of reagents. This excludes clinical material. Samples must be submitted as high quality extracted DNA, minimum 100uL. Turn-around time will depend on the number of samples and the queue of samples upon receipt.


Viral load - batch of 8 set-up - $5,357/Year

The rate is annually per sample virus. This is a set-up fee, which is due annually for each new virus that the core will quantitate using rate 1. This set-up fee covers necessary regulatory, quality-control, validation and data set-up work. The Vironomics Core Facility will maintain an infrastructure that is compliant with these regulations and perform the administrative action needed to be in compliance. These may differ depending on the particular virus.


96 gene viral array (clinical) - $871.00/Sample

The rate is per sample and provides for the sample genomic isolation, cDNA preparation, and analysis of this sample with 96 different primers using in-house developed arrays (IP contributed by the Dittmer group). This rate applies to clinical samples. Clinical samples require a heavy investment to conform with federal standards that apply to biosafety, GLP, HIPPA and GCLP regulations. Note that the core does not accept patient samples, or any samples that can be linked to individual persons. All samples must be de-identified. Research only, information cannot be used to treat or diagnose patients.

The rate is per sample and provides for the analysis of this sample with 96 different primers using in-house developed arrays (IP contributed by the Dittmer group). This rate only applies to non-clinical samples. The Core Facility will provide microarray services based on real-time QPCR. At this point the core is offering the following UNC-developed arrays: KSHV, EBV, HCMV, HSV-1, HSV-2, RRV, Viral Load, NFkappaB, Pre-miRNA, and P52response. This technology is more sensitive, more specific and targeted to specific needs of the user. This is in contrast to hybridization-based microarray facilities, which only use commercial assays to profile gene expression.
