The main goal of the Vector facility is development and implementation of viral vectors and viral vector-based techniques, which allow manipulating neural circuits and characterizing neural populations in laboratory animals. We also see the Vector facility as a pole of shared resources and expertise.
Many systems of recombinant viruses have been used in a wide number of applications, namely as gene delivery tools. Recombinant adeno-associated virus (rAAV) is very frequently a gene delivery vector of choice, especially in the field of gene therapy.
Our facility routinely produces batches (100-200 µl) of rAAV of serotypes 1 and 2 with high titer. Serotypes 5 and 9 can be also produced on a special request. Although the vast majority of produced rAAV batches have titers higher than 10E13 GC/ml, we guarantee a 10E12 GC/ml titer for any single rAAV production.
Virus quantification is performed by a qRT-PCR technique.
Typically, we produce one batch of a requested rAAV in a 2-4 weeks period for the price starting at 1000 USD. Some proteins, for example opsins, require more starting material to produce the same titer as a standard rAAV and this increases the cost price. Please note, that shipping costs are not included in the price.
To be able to produce rAAV accordingly to the above described characteristics we ask researchers to provide the desired gene construct in a pAAV backbone vector. If necessary, we offer assistance in subcloning into pAAV.
