The Genome Editing & Animal Models Core is committed to serving your genome-editing needs by providing expert services in consultation, design, reagent preparation and purification. We work with core facility partners on campus to deliver you genome edited mouse, rat, swine, and cell models (maize, soybean, and sorguhm coming soon). As a full-service genome editing facility open since 2015, TGF can design your project, and deliver your genotyped model. History of TGF: TGF was established to serve a strong, grass-roots need on campus. In February 2014, UW-Madison held a campus-wide workshop with the goal of bringing together those interested in CRISPR/Cas9. As interested scientists poured through the entrances and packed the aisles of the Genetics/Biotechnology Center Auditorium, it was clear there was a vigorous demand for genome editing resources on campus. The group organizing the workshop came to be called GEEWisc. Subsequent GEEWisc meetings sought to determine the types of campus resources that could best address genome editing needs across campus. A resoundingly strong request was for a core facility offering CRISPR expertise and delivering genome-editing services. To meet this need, TGF was established at the UW-Madison Biotechnology Center in March 2015, directed by Dr. C. Dustin Rubinstein. Today, TGF serves a diverse array of clients across campus and beyond. While the most vigorous demand is for the generation of genome-edited mice and rats in partnership with the Transgenic Animal Facility, the TGF is also developing swine models, cell line models, bovine models, and zebrafish models.

We will share our extensive CRISPR/Cas9 experience to guide your project from conception to the creation of your desired genome-edited model. TGF interfaces with UW-Madison facilities to generate your genome edited model. We have routinely and successfully generated many types of genome edits in many models, and TGF can provide genome-edited mice and rats, iPS cells, and plants. We will help you build your desired model of any species, but we specialize in the generation of mouse and rat CRISPR models. *Consultation* TGF can help you plan your genome editing project. Whether you are an experienced user of genome editing strategies or you simply have an idea that would allow you to ideally address your experimental questions, TGF is happy to provide ideas and suggestions on strategies and best-practices. *Design* TGF staff has extensive experience genome-editing projects. Any one of many design pitfalls can seriously imperil a genome editing project, and we are happy to help guide you through this critical stage. We have designed numerous mouse, rat, Drosophila, and cell-line genome editing projects. *Reagent synthesis* We have implemented a highly efficient, synthesis-based method for generating gRNA. We reliably generate purified, injection-grade gRNAs for rodent embryo microinjection. We also synthesize gRNA expression vectors, donor constructs, single-stranded donor molecules, and other reagents for genome editing projects. *Genotyping* We provide a number of genotyping solutions most appropriate for your model of choice. We offer high-throughput cell line screening for CRISPR/Cas9 generated knock-out projects. We also regularly genotype F1 litters for mouse and rat models.
