Kinetic and endpoint assay for extracellular ATP release from cultured cells with the sensitivity at low nM range.
Many cells release their ATP into extracellular space in response to various triggers, such as microbial toxins, mechanical stimulation, apoptosis, etc. Extracellular ATP (eATP) and its metabolites have profound effects on migration and activation of leukocytes, inflammation, and epithelial and endothelial barriers during infection. Using a luminescence-based assay we can measure eATP in real time for kinetic studies or at predetermined time points. We have shown eATP release from human lung epithelial cells after exposure to a pore-forming toxin from S. aureus (Lizak & Yarovinsky, Cell Host Microbe. 2012 Jan 19; 11(1): 70–80).
