Quantitation of Lentivirus Titer
Lentivirus titers can be measured in multiple ways. Viral suface protein measurements by ELISAs overestimate titer levels because it detects both active and dead virus particles. SBI measures virus titer by transducing target cells at several multiplicity of infections (MOIs) with subsequent qPCR analysis to determine the exact number of lentiviral integrations per target cell genome. This calculated titer is the most accurate method of measuring active virus particles and is reported as Infectious Units (IFU) per mL.

Exosomes are 60 - 150 nm membrane vesicles secreted by most cell types in vivo and in vitro. They are found in blood, urine, amniotic fluid, malignant ascite fluids and contain distinct subsets of microRNAs, mRNAs, lncRNAs and other ncRNAs. The identity and abundance of these RNAs has been shown to be a valuable approach to discover sequence signatures for the diagnosis and prognosis of disease. SBI announces our next-generation exosome RNA sequencing service to accelerate your biomarker discoveries. SBI's Exo-NGS service provides a complete start-to-finish solution from any biofluid to analyzed and aligned RNA-Seq data. We isolate the exosomes, purify the exoRNA and build the Illumina NGS libraries. These libraries are then sequenced using a 2x 75bp paired end Illumina NGS runs to provide greater confidence of RNA sequence identity.

Fully leverage the powerful nature of CRISPR/Cas9 and TALEN genome engineering platforms by using matching HR vectors. The majority of genome editing events affect only a percentage of the total population of transfected cells, estimated at 1-80% for mono- or bi-allelic modifications depending on the platform, the cell type, and the DNA target of interest. Due to the wide variation in activity, performing downstream phenotypic assays in a background of wild-type cells is challenging, especially if the phenotype in question is subtle or difficult to discern. Therefore, tools in the form of donor or targeting vectors that contain 1) fluorescent or antibiotic selection markers and 2) a gene fragment of interest to knock-in, knock-out, or correct a wild-type sequence will be extremely useful for obtaining a homogeneous population of cells whose genomes have been successfully targeted. SBI has generated a suite of homologous recombination targeting vectors suitable for the purpose of engineering both protein coding and non-coding genes including microRNAs and LncRNAs genomic loci.
SBI routinely generates high quality exosome peptide libraries for Mass spec analysis. The Mass Spec service works with exosomes isolated using ultracentrifugation as well as using ExoQuick (serum, plasma, ascites samples) or ExoQuick-TC (cell media, urine, spinal fluid) or immunopurified for specific exosome subpopulations using SBI's Exo-Flow IP kits. The MS service offers the choice of creating either exosome surface protein "shaving" peptide libraries for surface marker discovery or complete exosome peptide library preparations. The peptide libraries are then analyzed by nano LC/MS/MS with a Waters NanoAcquity HPLC system interfaced to a ThermoFisher Q Exactive. Peptides were loaded on a trapping column and eluted over a 75um analytical column at 350nL/min using a 2hr reverse phase gradient; both columns were packed with Jupiter Proteo resin (Phenomenex). The injection volume was 30uL. The mass spectrometer was operated in datadependent mode, with the Orbitrap operating at 60,000 FWHM and 17,500 FWHM for MS and MS/MS respectively. The fifteen most abundant ions were selected for MS/MS and data analyzed using MASCOT databases and Scaffold software.
SBI performs custom particle characterization services using the NanoSight technology to visualize, measure and characterize virtually all exosomes and microvesicles. Exosome particle size and concentration can all be analyzed simultaneously. Obtain accurate exosome concentration and size measurements all in one report.
Have SBI isolate and characterize exosomes from your biofluid samples (serum, urine, media, CSF, etc.). You can also send SBI exosomes you have already isolated and we can perform the nanoparticle tracking analysis. Each sample will be gently vortexed at 2.5k for 10 sec and then bath sonicated for 10 min at 33°C to ensure adequate exosome dispersion in the solution. NanoSight measurements are carried out in 0.02 um filtered PBS to remove any background and then visualized on an LM10 NanoSight instrument.
