SOLVE is a contract analytical lab, helping customers all around the world with AF4-MALS, SEC-MALS, AUC and DLS analysis and method development. 

 

Our expertise lies in the characterization of materials at the nano- and microscale. One of our core technologies – field-flow fractionation, AF4, FFF – is listed as a preferred method for investigating/validating protein aggregation by the US food and drug administration (FDA), emphasizing the push for more comprehensive characterization strategies.

FFF-MALS

Field-flow fractionation with multi-angle light scattering

A typical use of asymmetric flow field-flow fractionation (AF4) at SOLVE is for size and molar mass determination. It is applicable to samples with size above ~3 nm / 10 kDa. AF4 is also applicable to aggregate analysis and to the analysis of large polydisperse compounds with tendency to aggregate or adsorb to surfaces. AF4 is as an orthogonal technique to size exclusion chromatography (SEC). One recent application of AF4 is the analysis of protein and antibodies in complex matrixes such as serum and plasma. At SOLVE we use AF4 instrumentation from Wyatt Technology, coupled to UV, RI, MALS, and FL detectors.

Applicability:

AF4 is a method applicable to a very wide range of compounds, such as proteins, nanoparticles, viruses, and polymers. AF4 is applicable to compounds with molecular weight of ~ 10 000 g/mol and upwards to compounds of approximately 1 µm in size. Typical uses are for stability testing of proteins, aggregation analysis, batch to batch control, size and molar mass distribution.

Sample type: peptides, proteins, antibodies, liposomes, nanoparticles, viruses like particles, vaccine, polymer, and polysaccharides.

Sample requirements: Dispersed sample, minimum 100 ug sample.

Size exclusion chromatography with multiangle light scattering detectors

A typical use of size exclusion chromatography (SEC) at SOLVE is as a high resolution quantitative method for monitoring degradation and oligomerization. Our SEC instrumentation is connected to MALS-RI detectors to enable direct and reliable molar mass determination, as an alternative to molar mass determination with standards. We have columns from different manufacturers to cover the size range from peptides to large polymers. We also use UV and FL detection.

Applicability: Size exclusion chromatography (SEC) is applicable to a wide range of compounds, such as peptides, proteins, and polymers, with molecular weight of ~ 1000 g/mol up to ~10·106 g/mol. Typical uses are for stability testing of proteins, aggregation analysis, batch to batch control, molar mass distribution.

Analytical Ultracentrifugation (AUC)

We primarily use analytical ultra centrifugation (AUC) for validation purposes, and for the analysis of especially challenging samples after other characterization techniques had been tried (SEC and AF4). We operate AUC instrumentation with a wide range of rotors and cells, where the sedimentation of components is monitored by UV and interference optics (RI).

Dynamic Light Scattering

A typical use of dynamic light scattering (DLS) at SOLVE is particle size analysis and screening of protein solutions for aggregates. We have the traditional DLS cuvette format, but also a DLS plate reader for high-throughput analysis and screenings, that utilizes low sample volumes. Our instrumentation is temperature controlled which is handy for stability investigations. We use DLS instrumentation from Wyatt Technology and Malvern. Applicability: Dynamic light scattering (DLS) is applicable to particles and macromolecules from ~ 1 nm to ~1 µm. It can thereby be applied to everything from antibody, peptides and proteins, to nanoparticles and viruses. Typical applications of DLS are particle size analysis, screening of buffer conditions during formulation, stability testing, and aggregation analysis.
