T cells can be activated in vitro with anti-CD3/anti-CD28, PMA/Ionomycin or Super Antigen stimulation. Alternatively, T cells can be co-cultured with DCs and stimulated in an antigen-specific manner (e.g. Tetanus toxoid).
Read-out includes proliferation with 3H thymidine or fluorescent proliferation dyes, as well as cytokine production and expression of activation markers.
This human whole blood basophil activation assay is well suited for small sample sizes. The basophils are activated in a physiological environment, and the flow cytometry read-out ensures quick and accurate results.
After isolation and expansion of regulatory T cells, their capacity to suppress proliferation of conventional T cells can be analyzed within allogeneic PBMC cultures. The suppression of CD4+ and CD8+ T cell proliferation can be measured by flow cytometry.
This method of quantification enables analysis of up to 30 different cytokines, chemokines or growth factors in a single sample.

- Our laboratory is GMP and ISO 15189 certified
- MAT is executed according to European Pharmacopeia chapter 2.6.30 (Method A, B and/or C)
- Potential pyrogen contamination will be quantified relative to an international standard of endotoxin
- Experienced in MAT testing of proteins and antibodies from human and recombinant origin
Based on customer needs we perform product-specific validations. We also provide MAT feasibility testing, method development, SOP writing and in-process/batch release testing to reduce the workload of your laboratory. The customer will receive either a result letter, for in-process/batch release testing, or a (validation) report describing the performed tests and the obtained results.

Besides the NK cell cytotoxicity assays, we have several assay set-ups available to determine activation, proliferation and differentiation of immune cells, mainly using flow cytometry as a read-out. We generally focus on complete peripheral blood mononuclear cells or isolated T cells for these type of analyses, but these can of course also be applied to NK cells.
This human whole blood B cell activation assay is well suited for small sample sizes. The B cells are activated in a physiological environment, and the flow cytometry read-out ensures quick and accurate results.
This assay can be performed as a one-way MLR by co-culturing irradiated PBMCs from one donor with untreated PBMCs from another donor, or as a two-way MLR through co-culture of PBMCs from two untreated donors. It is also possible to isolate T cells from a donor before co-culture and use those for read out of proliferation. Proliferation can be monitored through incorporation of 3H thymidine, or, when more detailed information is required, can be measured by combining a FACS staining with a fluorescent proliferation dye.
We have a state-of-the-art flow cytometry facility and can offer fully customizable, elaborate multi-colour flow cytometry panels to allow in-depth phenotyping of any type of immune cell.
