Renovo Neural, Inc. is non-clinical contract research organization. We are a spin-out of the Cleveland Clinic, with expertise in translational research for degenerative neurological disorders. We offer unique cell-based assays as well as in vivo models for research in Multiple Sclerosis, Parkinson’s Disease, and other neuro-degenerative diseases. Related support services include MRI for rodent models, histology, immunohistochemistry (IHC), and related quantitative analysis. In addition, we offer 3-D electron microscopy for CNS, optic, and bone samples, and assessment of sub-cellular structures such as membranes, mitochondria, and other organelles.

Utilizing a technique called serial block-face scanning electron microscopy (SBFSEM) provides cut, imaged, and processed ultrastructural data at a fraction of the time of traditional methods. This generates a cost-effective, aligned stack of hundreds of serial sections overnight at nanoscale resolution. Renovo Neural is a two-system operation which allows for quicker turnaround time as well as expanded microscope capabilities. This service includes staining and imaging, standard and custom analysis (including g-ratios, internodal length, etc.), and reconstruction. The 3D electron microscopy team has experience imaging a wide variety of tissue, including nerve, cardiac, bone, and liver.

Rodent MRI studies on preclinical animal models of neurodegenerative diseases can increase the efficiency of studies. Renovo offers a range of readouts including anatomical MRI, MT weighted imaging, diffusion tensor imaging, manganese enhanced-MRI, contrast based imaging, functional MRI, and pharmacological MRI.

Multiple Sclerosis Mouse Model for Experimental Autoimmune Encephalomyelitis ('EAE')

The EAE is a common model for preliminary assessment of potential therapeutic compounds for Multiple Sclerosis. The EAE model induces inflammatory T-cell and demyelinating auto-antibody responses. Therapeutic compounds may be administered on a prophylactic basis or to assess therapeutic potential after disease onset. A variety of histological and immunohistochemistry endpoints are available.

Multiple Sclerosis Mouse Model for Neuroprotection, Demyelination, and Remyelination

Renovo’s Modified Cuprizone remyelination / neuroprotection model shows consistent levels of axon demyelination in the mouse, This provides an excellent platform for qualitative and quantitative readouts of remyelination or neuroprotective potential for therapeutic drug candidates in a mouse model.
In-life data collections can include behavioral assessments and / or MRI across multiple timepoints in studies from 6 weeks to 18 weeks duration (or beyond). Common endpoints include histology and immunohistochemistry of corpus callosum and other CNS tissues of interest to the Sponsor.

Among our histology offerings are tissue processing (animal perfusion , tissue fixation, and epon, paraffin, frozen, or plastic embedding), tissue sectioning (freezing microtome, cryosta, microtome, ultramicrotome semi-thin, free floating, and slide mounted sections), standard and specialized staining (Nissl, H&E, Tol Blue, Silver Staining, etc. as well as specialized stains to visualize features in the CNS and PNS tissue), immunohistochemistry (PLP, Iba1, GFAP, MAP2, etc. as well as customized IHC), imaging (bright field, epifluorescence, apotome, automated digital full slide, and confocal imaging), and analysis (qualitative and quantitative by automated or semi-automated techniques). Additionally, we aid the experiment design process and ultrastructural morphometrics.

in vitro Myelination Study on Synthetic Nanofiber Substrate

Renovo offers a unique assay to evaluate the capability of Sponsor’s test compounds to promote myelination upon a patented nanofiber substrate. Immuno-purified mouse OPCs differentiate into mature oligodendrocytes to form multi-level layers of myelin wraps on nanofibers. The assays provide visual and quantitative assessment of the amount of myelin produced, as well as the length of myelination upon the synthetic substrate.
Endpoint determinations are based on both the total number of myelinating cells plus the total length of myelinated nanofibers. Addition endpoints are optional, including PLP immunostaining.

Renovo offers cell-based assays to identify and evaluate compounds that stimulate OPC differentiation for potential remyelination therapies for Multiple Sclerosis (MS) and other myelin-related diseases. Our automated quantitative image analyses allow relatively high throughput primary and secondary screening of compound libraries to identify lead candidates. Renovo evaluates the ability of compounds to stimulate OPC differentiation using optimized immunohistochemical markers quantifying different stages of OPC differentiation.
