MST assays measure the interactions directly in solution without the need of immobilization to a surface (immobilization-free technology). MST uses a small amount of samples and allows for quick assay development and affinity determination (KD) of interactions. Hollow Fiber Assay

Selection of suitable cell lines for in vivo efficacy studies.

The Hollow Fiber Assay is suited to investigate the impact of a drug on three cell lines simultaneously in one mouse.

Approach:
The fibers are loaded with a suspension of tumor cells and get closed on both ends. Fibers of three cell lines get implanted subcutaneously and/or intraperitoneally into one mouse. The mouse will than be treated with the test substance for several weeks. At the end of the study, the fibers get isolated from the mouse body and the number of living cells will be quantified.

This is a fast and economical approach to select suitable cell lines for in vivo studies to investigate the impact of a drug on the growth of tumors.

Metastatic Tumor Animal Models

Metastasis tumor models are designed to analyze the anti-metastatic effects of anti-cancer drug candidates.

Syngeneic and xenograft models are available with intravenous, intracardial or orthotopic implantation.

Approach: Luciferase-transduced tumor cells with metastatic potential are implanted in mice. Metastasis is monitored via whole body imaging during the treatment period. After termination of the study, tumor cells will be quantified in a variety of organs such a lung, brain, liver or spleen via ex vivo luciferase analysis.

Bone marrow engraftment of leukemia cells will be performed after cyclophosphamide treatment.

Our InVivo Angiogenesis Model is suited for assessing the pro- or anti-angiogenic effects of drug candidates by analysing their impact on vessel formation of human endothelial cells in mice.

Approach:
Spheroids from human endothelial cells (HUVECs) are implanted subcutaneously in mice in a matrix containing angiogenic growth factors. The formation of a human vasculature can be monitored in vivo and ex vivo and the effect of treatment can be determined.

Compound Profiling in Cancer Cell Panels

Starting at $11,400.00 per sample

Our CL100-Prolifiler is a 2D proliferation assay suited for analysis of cytotoxic and anti-proliferative effects of test substances on a panel of 100 tumor cell lines.

Approach:
Tumor cells will be seeded in multiwell plates and incubated with the test substance for 3 days. After the incubation period, the amount of live cells will be quantified. This assay is usually performed for determination of IC50 values of the test substance.

Advantages:
Simple assay allowing for testing of large quantities of compounds in a rapid cost-efficient fashion.

We offer Free Choice Cell Proliferation Assays to choose a selection of tumor cell lines from our panel of 130 established cell lines.

On a quarterly basis we screen our entire panel of tumor cells lines. Any customer is welcome to provide one or several test items. Because all items will be tested in one run, this is an economic approach for a comprehensive overview of the efficacy of the test substance in a variety of tumor cells from different origins.

Colony Formation Assays

Synonymous for Soft Agar Assay or Clonogenic Assay

This assay is suited to investigate the impact of a drug on anchorage-independent growth of tumor cells.

Approach:
A single cell suspension will get mixed with soft agar and pipetted into a 96-well plate. The soft agar hardens making the cells float - not touching the plastic.
The drug will be applied to the wells and incubated for up to three weeks. The tumor cells proliferate and the single cells start forming colonies. At the end of the incubation period, the amount of cells per well will be quantified.

3D Tumor Models

Tumor spheroids are used to screen compounds in a near-physiological tumor situation.

Monospheroids are available for testing as well as co-culture spheroids assembled from tumor and stroma cells.

Approach:
Spheroids autonomously assemble in U-bottom 96-well plates. Incubation of compounds for three days with subsequent quantification of viable cells.

In co-culture spheroids the amount of stroma as well as tumor cells will be measured simultaneously via detection of renilla luciferase (stroma cells) and firefly luciferase (tumor cells).

Oris assay to measure the anti-migratory effects of test substances on tumor cells.

Approach:
Tumor cells are seeded in a 96 well dish containing rubber stoppers. After cells reach confluency the stoppers will be removed, the test substance will be added to the medium and cells are allowed to migrate into the center of the dish. The amount of migrating cells will be determined by fluorescent labelling to determine the anti-migratory effect of the test substance.

In vitro Angiogenesis Assays

Assay for determination of anti- or pro-angiogenic effects of test substances on vessel formation by endothelial cells.

Approach:
Human endothelial cells (fibroblasts or smooth muscle cells may also be used) form spheroids by aggregation in a hanging drop system. The spheroids will be transferred into a gel-like matrix containing the test substance. The gel-like matrix allows the spheroids to float and vessels start sprouting into all directions. After 24 hours, the amount of newly formed vessels will be quantified to determine the anti- or pro-angiogenic effect of the test substance.

Plasma Inhibitory Activity (PIA) Assay

This assay is suited to investigate the impact of plasma on the activity of a kinase inhibitor.

Usually kinase inhibitors in cellular assay systems incubate in serum-free medium. When this drug is applied to a mouse or a human it will face high amounts of plasma, which potentially bind to the compound and decrease its activity.

Approach: We established two ways to investigate the activity of kinase inhibitors in the presence of plasma: incubation of the drug with plasma before investigating its kinase inhibitor activity in a cellular test system. The second possibility is to inject the drug into mice. Blood will be drawn which contains the drug which than will be tested for its kinase inhibitory activity in a cellular test system.

Both tests can be used in comparison with inhibitors that are known to be inhibited by plasma and are unaffected by surrounding plasma.

IC50 determination of the kinase inhibitory activity will be performed to determine the amount of freely-availalbe kinase inhibitor.
