Proteos offers production of full-length IgG antibody, F(ab) and (Fab’)2 antibody fragments, and antibody fusion proteins. Our antibody production capabilities include construct design, transient mammalian cell expression, utilizing in-house or customer provided expression vectors, and affinity purification. Additional purification services offered include size exclusion chromatography of up to 5 grams of antibody in a single production run, buffer exchange by dialysis, endotoxin testing, analytical SEC, SDS-PAGE (reducing and non-reducing), and sterility testing.

Our industry leading transient expression platform utilizes fully licensed HEK (human embryonic kidney) and CHO (Chinese hamster ovary) cells lines expressing the Epstein-Barr virus (EBV) nuclear antigen 1, with proprietary expression vectors containing the EBV origin of replication (OriP) to allow episomal plasmid amplification and high level recombinant protein expression.

We utilize optimized and fully validated mammalian cell expression protocols for consistent, reproducible protein expression in serum-free shake flask culture volumes ranging from 30 mL to 60 L per batch. Only mycoplasma-free, high viability, low passage cultures are used for transfection and are monitored using a Beckman Coulter Vi-cell XR for culture density, viability, and average cell diameter. The Vi-cell XR uses the trypan blue dye exclusion method and averages 50 measurements to compute the parameters for each sample. This allows us to monitor cultures with a high degree of accuracy which is imperative for the quality and reproducibility of protein production projects. All cell culture data is included in a customized technical report delivered with each project.

Proteos offers baculovirus mediated protein expression in our fully licensed Sf9 (Spodoptera frugiperda) and Tni PRO (Trichoplusia ni) cell lines. Both cells lines are suitable for the production of intracellular or secreted recombinant proteins with a majority of the post-translational modifications found in mammalian cells. The Tni PRO cell line has been shown to produce higher levels of secreted protein expression in many cases.

The baculovirus expression vector system (BEVS) uses homologous recombination or site-specific transposition to produce recombinant virus encoding the protein of interest. To ensure accurate MOIs (multiplicity of infection) and reproducibility of each production run, Proteos utilizes a robust plate-based flow cytometry assay for determining the titer of baculovirus stocks prior to infection of insect cells for protein production. The titer of each virus stock that we produce is determined in triplicate using our plate-based flow cytometry assay. We routinely produce high-titer virus stocks with titers ranging from 5 x 10^8 – 2 x 10^9 infection units (IU) per milliliter in a single amplification.

We utilize optimized and fully validated insect cell expression protocols for reproducible high-level protein expression in serum-free shake flask culture volumes up to 40 L per batch. Only mycoplasma-free, high viability, low passage cultures are used for expression and are monitored using a Beckman Coulter Vi-CELL ™ XR for culture density, viability, and average cell diameter. The Vi-CELL ™ XR uses the trypan blue dye exclusion method and averages 50 measurements to compute the parameters for each sample. This allows us to monitor cultures with a high degree of accuracy, which is imperative for quality and reproducibility of protein production projects. The thermal shift analysis (TSA) is a thermal denaturation assay for elucidating conditions that stabilize a protein in solution. Protein destabilization and melting is measured as an increase in fluorescence as hydrophobic binding sites within the protein become exposed during unfolding and are able to bind a fluorescent dye in the assay mixture. The melting temperature (Tm) of a protein is identified as the temperature at which 50% of the protein is unfolded. The assay is performed in plate format, allowing a large number of conditions (buffers, ligands, etc.) to be screened simultaneously. Conditions that cause the Tm to shift to a higher temperature are identified as stabilizing.

Proteos utilizes a robust economical plate-based flow cytometry assay for baculovirus titer determinations, allowing accurate optimization of infection parameters used for protein production in baculovirus infected insect cell cultures.

The titer (infectious units (IU) per milliliter) of a baculovirus stock is determined by measuring the expression of gp64 (a baculovirus envelope protein) on the surface of infected insect cells eight hours post infection (hpi).
Unlike plaque and end-point dilution assays that are laborious and often add weeks to a project; our method is quick, reliable and economical. Results can be obtained in as little as 3 business days from receipt of the samples, only requires 600 µL of baculovirus stock, and is performed in triplicate so you can be confident of the results. You will receive a full data sheet containing all of the assay data.
