The Proteomics and Metabolomics Core Facility offers a wide variety of services for separation, identification, quantification, and post-translational modification analysis. Results are usually delivered by email or discussed in person.
LC-MS/MS protein identification includes sample preparation, denaturation, reduction/alkylation, tryptic digestion, and mass spectrometry analysis. Typically, 90 minute gradients are used for peptide separation with MS and MS/MS in ion trap (LTQ) or orbital ion trap (LTQ-Orbitrap) instruments.
Data are reported in Scaffold files, and instructions are provided for data interpretation and selection of high confidence results. The free Scaffold viewer is required, and it can be downloaded from Proteome Software.
Discovery/Untargeted/Targeted Metabolomics/Lipidomics/Fatty Acid, HILIC/Reversed Phase UPLC separation, positive/negative mass spectrometry, metabolite identification and quantitation with library searching
Liquid Chromatography-Multiple Reaction Monitoring Mass Spectrometry (LC-MRM) is used to quantify components in complex mixtures. Our laboratory uses this technique for quantification of protein biomarkers. Assay development prior to sample analysis includes synthesis and characterization of standard peptides. Note that multiple proteins can be quantified in each sample.
