FACILITY OPENS 15 June 2020.
The Proteomics and Mass Spectrometry Facility is equipped with an ThermoScientific Orbitrap Elite mass spectrometer for high resolution and high mass accuracy analysis. It is coupled with a nano HPLC, increasing its capacity to analyze more complex protein mixtures. The facility also has a Bruker Autoflex MALDI for quick analysis of tryptic digests of pure proteins. The facility offers in-gel digestion and subsequence analysis for protein identification. Samples need to be ready for analysis as facility does not include any sample clean up. Analysis is performed on a Bruker Esquire 3000 Plus ion trap. MS/MS can be requested at time of analysis of a specified ion at no extra cost. A mass spectrum can be sent via e-mail as a emf (picture) file and/or x,y coordinate file for plotting with software such as Excel. Turn around time is general less than 3 business days.
High Resolution Accurate Mass ESI by sample infusion. Only manipulation of sample would be for dilution or solvation. Analysis will be performed on one of the following:Bruker Impact II QTOF ($50), Thermo Scientific Velos Orbitrap mass spectrometer ($55), Bruker SolariX 12T FTICR MS ($60).
Protein ID by Peptide Mass Fingerprinting (in-gel digestion, MALDI-MS, & database search). This is by TOF-MS and is good for ID of single protein in clean gel band. Performed on a Bruker Autoflex.
Protein ID of gel band by LC-MS/MS (60 min. run) (includes: in-gel trypsin digestion, LC-MS/MS, & one database search) Good for searching band(s) that are suspected to contain numbers proteins. Thermo Scientific Velos Orbitrap mass spectrometer coupled to a nano-LC system.
Sample is analyzed by a Bruker Autoflex MALDI-TOF. Samples are routinely mixed with DHB matrix for analysis but facility also has alpha-cyano-4-hydroxycinnamic acid and sinapinic acid matrices available. Data is provided as an emf (picture) file and/or x,y coordinates that can be plotted using Excel.
