The Protein Chemistry Technology Core was established to provide efficient, high quality services to researchers in the academic community.

The services presently offered include peptide synthesis and Edman protein sequencing. Our facility, its state-of-the-art instrumentation, and computer software is constantly upgraded to ensure the highest possible level of support. It is our aim to provide quality performance and offer experienced support, both through our products and people.

The ABI 494 protein sequencer can handle samples at 50-100 picomole levels, either attached to PVDF membranes or in solution. If in solution, please contact our lab for information on acceptable buffers and solvents. We can usually return the results within a few days.

Matrix-assisted laser desorption ionization time-of-flight mass spectrometry

The molecular weight of synthetic or naturally isolated peptides and small proteins (less than 5000 Da) can be measured by MALDI TOF mass spectrometry. The mass accuracy of the molecular weight determination will vary between ~10 ppm to 0.01% depending on the size of the molecule being measured.

Peptide synthesis at scales 25-250 micromole using Fmoc or t-Boc chemistry. Convergent synthesis using native chemical ligation. Wide variety of modifications including, phosphorylation, acetylation, methylation, fluorophores, stable isotopes, biotin, gold for Electron Microscopy etc.


Major instrumentation:

  • Applied Biosystems 433A: Single-peptide, batch synthesizer with Fmoc and t-Boc chemistry capability at a scale of 0.1 to 0.5 mmol.
  • Applied Biosystems 430A: Single-peptide, batch synthesizer with Fmoc and t-Boc chemistry capability at a scale of 0.1 to 0.5 mmol. Fast cycles allow a 20mer peptide to be synthesized in 8h.
  • Perseptive Biosystems Pioneer: Fmoc chemistry only, continuous flow instrument, which will synthesize two peptides simultaneously.
  • Rainin Symphony: Multiple-peptide synthesizer using batch-synthesis methodology and either Fmoc or t-Boc chemistry. Up to 12 peptides can be synthesized simultaneously.
  • Advanced Chemtech APEX396: Multiple-peptide synthesizer using batch-synthesis methodology and either Fmoc or t-Boc chemistry. Number of peptides that can be synthesized at one time depend on the RV block used and the desired synthesis scale.
  • Peptide Institute Hydrogen Fluoride Apparatus: Permits safe handling of HF required for final cleavage and deprotection of t-Boc synthesized peptides.
