The Protein Chemistry Technology Core was established to provide efficient, high quality services to researchers in the academic community.
The services presently offered include peptide synthesis and Edman protein sequencing. Our facility, its state-of-the-art instrumentation, and computer software is constantly upgraded to ensure the highest possible level of support. It is our aim to provide quality performance and offer experienced support, both through our products and people.
The ABI 494 protein sequencer can handle samples at 50-100 picomole levels, either attached to PVDF membranes or in solution. If in solution, please contact our lab for information on acceptable buffers and solvents. We can usually return the results within a few days.
The molecular weight of synthetic or naturally isolated peptides and small proteins (less than 5000 Da) can be measured by MALDI TOF mass spectrometry. The mass accuracy of the molecular weight determination will vary between ~10 ppm to 0.01% depending on the size of the molecule being measured.
Speed of turnaround, typically 5 days for crude (>70%) and 6 days for RP-HPLC purified (95%) peptides.
Ability to synthesize very difficult eg amyloidogenic peptides. We can make long polypeptides up to 100 residues using proprietary chemical strategies.
Peptide synthesis at scales 25-250 micromole using Fmoc or t-Boc chemistry. Convergent synthesis using native chemical ligation. Wide variety of modifications including, phosphorylation, acetylation, methylation, fluorophores, stable isotopes, biotin, gold for Electron Microscopy etc.
