Selection of suitable cell lines for in vivo efficacy studies.

With the Hollow Fiber Assay we investigate the impact of a drug on three cell lines simultaneously in one mouse.
The fibers will get loaded with a suspension of tumor cells and closed. The fibers get implanted subcutaneously and/or intraperitoneally into the mouse - three fibers containing three cell lines in one mouse. The mouse will than be treated with the drug for several weeks. At the end of the study the fibers get isolated from the mouse body and the number of cells will be determined.

This is a fast and economical approach to select suitable cell lines for in vivo studies to investigate the impact of a drug on the growth of tumors.

Metastasis tumor models are designed to analyze the anti-metastasic effect of anti-cancer drug candidates.

Syngeneic and xenograft models are available with intravenous, intracardial or orthotopic implantation.

Approach: Luciferase-transduced tumor cells with metastatic potential are implanted in mice. Metastasis is monitored via whole body imaging during a treatment period of several weeks. After termination, tumor cells will be quantified in a variety of organs such a lung, brain, liver or spleen via ex vivo luciferase analysis.

Bone marrow engraftment of leukemia cells will be performed after cyclophosphamide treatment.

Syngeneic mouse models for efficacy testing of anti-cancer drugs.

Our immuno-oncology platform comprises a panel of syngeneic tumor models for testing of immune-modulating drugs. We offer models with subcutaneous and orthotopic implantation as well as metastasis models.

Our models are evaluated by treatment with anti-PD1, anti-PDL1 and anti-CTLA4 treatment regarding their tumor-growth inhibitory impact as well as their effects on tumor-infiltrating immune cells.

Flow cytometry will be used to characterize changes to the several immune cell subsets such as macrophages, myeloid derived suppressor cells, T cells, DCs and neutrophils.

Advantages:
Simple assay allowing for testing of large quantities of compounds in a rapid cost-efficient fashion.

We offer Free Choice Cell Proliferation Assays for our customers to choose the individual tumor cell lines to be tested.
On a quarterly basis we screen our entire panel of tumor cells lines. Any customer is welcome to provide one or several test items. Because all items will be testing in one run, this is a very economic approach for a comprehensive overview of the efficacy of the test item in a variety of tumor cells of different origins.

Human endothelial cells (fibroblasts or smooth muscle cells may also be used) form spheroids by aggregation in a hanging drop system. The spheroids will be transferred into a gel-like matrix containing the test substance. The gel-like matrix allows the spheroids to float and vessels start sprouting into all directions. After 24 hours, we will quantify the amount of newly formed vessels and determine the anti- or pro-angiogenic effect of the test substance.

Cellular phosphorylation assays will increase your knowledge on which one of your inhibitors still blocking your kinase of intersest in the cellular context.

Advantages:
Cell-based test systems offer the advantage of physiological concentrations of ATP and other cofactors at the site of action and the phosphorylation of physiological substrates.

Approach.
Cells are pre-incubated with test compounds to allow thorough target binding. Depending on the assay we might stimulate the kinase with its specific ligand. After 90 min incubation time, cells get lysed and the phosphorylation status of the specific substrate of the kinase will be determined via ELISA.
