Selection of suitable cell lines for in vivo efficacy studies.

The Hollow Fiber Assay is suited to investigate the impact of a drug on three cell lines simultaneously in one mouse.

Approach:
The fibers are loaded with a suspension of tumor cells and get closed on both ends. Fibers of three cell lines get implanted subcutaneously and/or intraperitoneally into one mouse. The mouse will than be treated with the test substance for several weeks. At the end of the study, the fibers get isolated from the mouse body and the number of living cells will be quantified.

This is a fast and economical approach to select suitable cell lines for in vivo studies to investigate the impact of a drug on the growth of tumors.

Metastasis tumor models are designed to analyze the anti-metastatic effects of anti-cancer drug candidates.

Syngeneic and xenograft models are available with intravenous, intracardial or orthotopic implantation.

Approach: Luciferase-transduced tumor cells with metastatic potential are implanted in mice. Metastasis is monitored via whole body imaging during the treatment period. After termination of the study, tumor cells will be quantified in a variety of organs such a lung, brain, liver or spleen via ex vivo luciferase analysis.

Bone marrow engraftment of leukemia cells will be performed after cyclophosphamide treatment.

Human endothelial cells (fibroblasts or smooth muscle cells may also be used) form spheroids by aggregation in a hanging drop system. The spheroids will be transferred into a gel-like matrix containing the test substance. The gel-like matrix allows the spheroids to float and vessels start sprouting into all directions. After 24 hours, the amount of newly formed vessels will be quantified to determine the anti- or pro-angiogenic effect of the test substance.

This assay is suited for testing of kinase inhibitors in a cellular setting.

In comparison to biochemical kinase testing, cell-based test systems offer the advantage of physiological ATP concentrations at the site of action, a membrane barrier to cross and physiological substrates.

Approach: Cells which express the kinase of interest will be preincubated with test substance to allow thorough target binding. In case of receptor tyrosine kinases we determine the level of autophosphorylation of this kinase. For serine/threonine kinases we developed assays that allow the quantification of the phosphorylation status of a downstream substrate.

This assay is suited to investigate the impact of plasma on the activity of a kinase inhibitor.

Usually kinase inhibitors in cellular assay systems incubate in serum-free medium. When this drug is applied to a mouse or a human it will face high amounts of plasma, which potentially bind to the compound and decrease its activity.

Approach: We established two ways to investigate the activity of kinase inhibitors in the presence of plasma: incubation of the drug with plasma before investigating its kinase inhibitor activity in a cellular test system. The second possibility is to inject the drug into mice. Blood will be drawn which contains the drug which than will be tested for its kinase inhibitory activity in a cellular test system.

Both tests can be used in comparison with inhibitors that are known to be inhibited by plasma and are unaffected by surrounding plasma.

IC50 determination of the kinase inhibitory activity will be performed to determine the amount of freely-availalbe kinase inhibitor.

1. The Free Choice Kinase Assay allows for characterization of the inhibitory capacity of your test substance against your choice from 430 kinases.
For each project you can freely define (i) the number of test compounds, (ii) the kinase panel, (iii) and the number of data points per test (i.e. number of concentrations and replicates).

  1. Selectivity profiling with a choice of two broad kinome panels:
    The Wild Type Profiler consists of 320 wild type protein kinases.
    The 410- Profiler contains the wild type protein kinases plus lipid kinases and disease-relevant mutants.

Protein kinases get screened with a radiometric activity assay using human kinases, [33P]-phosphate labelled ATP and mostly generic substrates. During the kinase reaction the 33P gets transferred onto the substrate which than will be quantified via scintillation counting.

Lipid kinases get screened with the ATP consumption assay which is also an activity assay.

Both technologies are suitable to investigate all classes of kinase inhibitors. Most kinases are full lenght proteins allowing the compound to bind at its individual site which can differ from the ATP binding site.

Small scale projects as well as high-throughput studies are possible.

1. The Free Choice Kinase Assay allows for characterization of your test compounds in a self-selected panel from 17 lipid kinases.
For each project you can freely define (i) the number of test compounds, (ii) the kinase panel, (iii) and the number of data points per test (i.e. number of concentrations and replicates).

  1. Selectivity profiling of 13 lipid kinases alongside 397 protein kinases with our 410-Profiler.

Assay technology:
We test your compounds against our lipid kinases with an ATP consumption assay. More specific, we use the ADP Glo assay from Promega which will quantify the amount of ATP that got converted into ADP during the kinase reaction. This assay is an activity assay and suitable for the investigation of every class of kinase inhibitor.

With our extensive experience of 20 years of kinase assay development and screening we are the right partner for the establishment of an assay for your kinase of interest.

We can either use a kinase that is commercially available or we can produce the kinase for you at our recombinant protein production site. Once we have the kinase in our hands, we will find a suitable substrate with our SubstrateFinder and examine the activity of the kinase with different substrate concentrations, ATP conditions and we might adjust co-factors or elements that increase the activity of the kinase if necessary.

Beside simple single-step assays we are also able to establish cascade-assays such as in the example available for download from our website:

Importantly, all of our off-the-shelf active kinase enzymes are used once a month in our profiling services, which serves as perfect quality control to guarantee their activity. We can provide you information about suitable substrates and assay conditions which is an extremely valuable resource for our customers. Our scientists will also help you troubleshoot your kinase assays.

We routinely test the activity of our kinases in our radioactive assay format but also in the ATP consumption assay ADP-Glo.
