Leaderna Biostructures offers a customized service for crystallization of protein-nucleic acid complexes – one of the most promising technologies for drug discovery and development. The approach allows to establish the detailed molecular mode of interaction between proteins and nucleic acids in cellular processes and diseases. With molecular interactions unveiled to the smallest detail, the service accelerates the design of drug-like molecules in DNA-like antisense and RNA-based drug therapies. Based on structural data, antisense oligonucleotides (ASOs) or dsRNAs specific for a particular target can be designed to maximize the potential of action with best efficiency and specificity. We have unique expertise in the field we've developed through cooperation with Ionis Pharmaceuticals since 2016. Thanks to the years of our mutual projects we’ve found a market niche to support companies in the development of nucleic acid-based drugs.

We offer two approaches of protein-nucleic acid crystallization with customization depending on the particular project requirements:

Standard approach - starting point for any new project; we perform co-crystallization of protein and nucleic acid, with optimization of stoichiometry of the two molecules. We use crystallization screens dedicated for nucleic acids to maximize the chance of getting suitable crystals.

Complex approach - for challenging projects; when standard co-crystallization fails, we introduce a unique chemical cross-linking technology. Both the protein and nucleic acid are modified that allows reversible site-specific tethering of the DNA/RNA molecule to the target protein. An advantage is the stabilization of transient and unstable protein-nucleic acid complexes, which facilitates the crystallization of otherwise non-crystallizable complexes.

X-ray crystallography is currently the most accurate technology for determining 3D structure of a protein. This type of analysis allows the researchers to determine the position of each of the thousands of atoms of the studied molecule with great precision and create a three-dimensional model. This makes crystallography one of the most advanced and promising approaches to new drugs discovery. High resolution of the protein structure, which is a potential goal for therapy, allows for the design of specifically tailored chemical molecules that can interact with it. We offer X-ray crystallography of protein, protein–ligand complexes as well highly specialized standalone service in Crystallization and structural analysis of protein-nucleic acid complexes.

Cryo-EM enables the determination of high-resolution structures of proteins under near native condition at close to atomic resolution and complements existing atomic-resolution approaches, such as X-ray crystallography. Recent publications show that cryo-EM can be used to investigate a broad spectrum of drug-target interactions with routine resolution of ~4 Å, possibly up to 2 Å. Cryo-EM enables the analysis of proteins or protein complexes with a molecular weight ≥150 kDa (e.g., antibodies and membrane proteins) that are otherwise difficult or impossible to analyze using other structural biology methods.

  • Negative stain EM – preliminary negative stain analysis
  • Negative stain EM – optimization of the sample homogeneity

Structural biology services - coupled cryo-EM/X-ray crystallography approach

Structural analysis of large macromolecular complexes remain a challenge. The fundamental attributes of perfect structure are high quality data on global conformational changes between and/or within individual subunits of the entire complex, while preserving the ability to detect discrete features like specific interactions with the ligand/drug at the binding site.

Two powerful methods of structure determination of biomolecules have been routinely applied to achieve one of these goals at a time. While single particle cryo-EM provides a lower resolution and more biologically relevant data of large macro-complexes, X-ray crystallography yields a high-resolution structural information on individual protein units and small molecules.

To reach the highest expectations, ProBiostructures recommends the coupled cryo-EM/X-ray approach which links and integrates the favorable features of both methods. Assays of protein secondary structure and its stability

Stabilization and formulation of therapeutic proteins against physical instability remain one of the top factors to achieve. The best indicator of stability is protein secondary structure measurement that immediately captures the influence of physical and chemical factors as well as modifications of protein sequences (substitutions, deletions and fusions) on structural integrity of a protein.

We offer three complementary methods that can be used independently in any combination:

  • CD (Circular Dichroism) spectroscopy - [Jasco J-810, Bio ATRII measurement cell]
  • FTIR (Fourier Transform Infrared) spectroscopy - [Bruker Tensor 27]
  • DSC (Differential Scanning Calorimetry) - [Microcal VP-DSC]

Quaternary structure and aggregation analysis

Very frequently particular macromolecule participates in several such assemblies and this property is crucial for its biological function. This is why structural studies of such assemblies by Cryo-EM and X-ray crystallography are at the forefront of deciphering most important biological processes at the molecular level. Biophysical methods are relatively fast, require less material and therefore might be useful to establish predominant class (and stoichiometry) of macromolecular assembly and conditions best stabilizing it. These methods are also excellent for confirming sample monodispersity and lack of aggregation that are prerequisite for successful structural studies. 


We offer two complementary methods that can be used independently:

  • AUC (analytical ultracentrifugation) - [ProteomeLab XL-I - Beckman]
  • SEC-MALS (Size Exclusion High Precision Liquid Chromatography with Multi Angle Light Scattering) - [HPLC - Waters; detectors Dawn 8+ [MALS] & Optilab T-rEX refractometer - Wyatt]

Molecular interactions analysis

Full characterization of interaction of a lead compound or prospective drug with its intended and unintended targets encompassing kinetics and thermodynamics parameters serves as a primary tool guiding drug development process. In basic research similar data are indispensable for in depth quantitative description of particular interaction.


We offer two complementary methods for label-free interaction analysis that provide characterization of molecular interaction from different angles with little overlap and a classical method that requires fluorescent label on the smaller interactant:
