A&G’s Precision Antibody™ employs a staff of 20 based in an 8,000-square-foot laboratory facility in Columbia, Maryland.
We offer several different methods of productions and levels of purification. In our basic IgG production/purification project, monoclonal antibody is purified on a Protein G affinity column using AKTA FPLC system. Purified antibody is tested for its antigen binding ability by solid phase EIA and the purity checked by SDS-PAGE. Protein concentration assessed by A280. Purified antibody is membrane sterilized. All of the purification methods, chromatogram, EIA data, Coomassie stained SDS-PAGE gel image and protein quantification data will be sent to our client with a Certificate of Analysis (COA).
We also offer additional services including endotoxin testing, sterility testing, size exclusion chromatography, isoelectric focusing, buffer exchange, protein quantification by micro BCA, IgG quantification by EIA, adjustment of IgG concentration to specification, vialing to specifications, and cell banking.

In Vitro Antibody Production
Precision Antibody has broad experience in pilot scale, small scale, and large scale mammalian cell culture of hybridomas for production of monoclonal antibodies. Our facility is equipped with tissue culture bags, roller bottles, bioreactors, hollow fiber systems, and stirred tank fermenters. Our Precision Antibody Team will help you choose the best and most cost effective method to produce your monoclonal antibodies.
Pilot Antibody Production (1 mg to 10mg)
Most of our pilot antibody production is done in disposable bioreactor bags. The benefits of flexible bag containers include faster set-up, reduction of down time, and simplified validation. Disposable bags also greatly reduce the risk of cross contamination. These bags are a perfect solution for pilot scale productions. Our Precision Antibody team passes on these benefits to our customers in the form of lower costs for 1mg to 10mg antibody productions.
Small Scale Antibody Production (10mg to 500mg)
Small scale antibody production of your desired monoclonal antibodies can be reliably achieved in roller bottles or disposable bioreactors. Yields of monoclonal antibodies in roller bottles and bioreactors are clone dependent and vary on the cell line's growth and antibody production rates. Precision Antibody helps customers refine their growing conditions by assisting customers in their choice of media formulation. Whether selecting low bovine IgG fetal bovine sera, serum-free media, or animal protein-free media, Precision Antibody will help you hone your antibody production and deliver the required amount of antibody for your application.
Large Scale Antibody Production (500 mg to gram quantities)
Precision Antibody uses Fibercell System and Stir Tank Fermenters for industrial scale productions. We work with customers to determine antibody yield and media conditions. If necessary, we perform a pilot production to assess the antibody production rate. If the cell line is a low producer, it may be advantageous to re-clone and identify higher immunoglobulin producing clones (extra charge). The FiberCell® Systems hollow fiber system is the method of choice for the in-vitro production of 100 mg to 1 gram or more of monoclonal antibody.
Our stirred Tank Fermenters are used for projects over a gram. Again, Precision Antibody helps customers refine their growing conditions by assisting customers in their choice of media formulation - low bovine IgG fetal bovine sera, serum-free media, or animal protein-free media. Precision Antibody will help you hone your antibody production and deliver the required amount of antibody for your application.
CHO, NSO and HEK-293 Cells
Besides our custom antibody monoclonal production, Precision Antibody has experience in antibody production for CHO, NSO and HEK-293 cells. We work with customers to determine cell line characteristics, media requirements, and purification needs.
In Vivo Production
Precision Antibody also has a nude mice facility for ascities production. Occasionally, we encounter hybridoma cells that do not produce monoclonal antibodies in in vitro systems. Ascites production in nude mice can be used as an alternative method to produce these monoclonal antibodies. Additionally, ascites production often yields higher concentrations of monoclonal antibody. We prefer to use nude athymic mice for ascites production to eliminate the contamination of endogenous mouse IgG.

Choosing the right antibody is critical to the success of your immunoassays. In the course of assay development, choosing the right antibodies with high affinity and low off rate at the start of assay development is critical. Biacore analysis examines the characteristics of your antibody—antigen interaction. It uses Surface Plasmon Resonance (SPR) technology for the label-free, real time measurement of association (on) & dissociation (off) rates (ka & kd) and binding affinity constant (KD) of your antibody antigen interaction. Incorporating Biacore scouting assays early in the antibody screening process allows you to rank your monoclonal antibodies based on their binding kinetics and help you select the right antibodies for your assay. A relative epitope scouting assay with Biacore SPR technology can easily identify antibodies that bind to distinct separate epitopes on your antigen. Biacore assay helps you establish a panel of monoclonal antibodies with known binding kinetics to different epitopes of the antigen. For sandwich immunoassay development, it is easy to identify a pair of antibodies that can be used as capture and detection antibodies. A full binding kinetic analysis can then be performed on your final assay candidate antibodies.
Biacore assay is not limited to the characterization of antibody antigen interactions. Biacore assay can be also used for determination of label-free binding and a real time analysis of the strength of any molecular interactions provided that the molecular mass can be detected by the instrument (molecular mass above 1,200). Antibody Scouting Analysis (ranking by affinity)
Biacore scouting analysis of antibodies are carried out using Biacore 3000 in order to rank the selected mAbs according to its KD calculations. Certificate grade CM5 chip are coated with capture antibody (goat anti-mouse IgG Fc for mouse mAbs, goat anti-human IgG Fc for chimeric, humanized or fully human antibody). The test mAb is then captured on the chip and analyte (antigen) at a saturating concentration will be passed through to measure rough ka and kd in real time. The KD will be determined from the ka and kd values obtained. Based on the kinetic parameters, test antibodies can be ranked according to their binding affinity and dissociation rate (off-rate or kd).
Full Binding Kinetic Analysis
Biacore full binding kinetic analysis of antibodies are carried out using Biacore 3000. Detailed binding kinetic parameters (association rate, ka, dissociation rate, kd, and affinity constant, KD) can be determined by full kinetic analysis. Certificate grade CM5 chips are coated with capture antibody (goat anti-mouse IgG Fc for mouse mABs, goat anti-human IgG Fc for chimeric, humananized or fully human antibody). The test mAb is then captured on the chip and the binding kinetics will be measured at up to 6 different analyte (antigen) concentrations (from 0 to saturating concentration). Binding kinetic parameters (ka, kd, & KD) at each analyte concentration will be measured and a χ2 analysis will be performed to assess the accuracy of the data. In our experience, high affinity antibodies with low off-rate exhibit good utility in immunoassays.
Relative Epitope Mapping
Biacore epitope scouting analysis of antibodies are carried out on Biacore™ 3000 for Epitope Mapping in order to identify mAbs that recognize distinct separate epitopes on a single antigen. Certificate grade CM5 chips are coated with capture antibody (goat anti-mouse IgG Fc for mouse mAbs, goat anti-human IgG Fc for chimeric, humanized or fully human antibody). In the first step, mAb is captured on the chip and analyte (antigen) will be passed through to allow binding. A second mAb will be applied to the Ab-Ag complex to examine binding. If the second antibody binds to a separate epitope, we expect to observe additional binding on the sensorgram.
Alternatively, scouting assays can be carried out by capturing the antigen on the chip and sequentially passing the antibodies over the antigen to observe antibody binding. By measuring antibody binding using combinations of test antibodies, antibodies binding to different epitopes and to the same epitope are identified.

At the end of the Biacore project, our team provides a sensorgram of antibody antigen interaction and a summary of the kinetic data (ka, kd, KD).

Precision Antibody will perform a titer check at day 21 and provide you the results. Typically, the titer on Precision Antibody’s 21 day rabbit is between 1:30,000~1;100,000. Titer is defined as serum dilution to achieve A620=0.15 where the control reading is A620=0.05 in EIA assay.
The terminal bleed can be performed at any time after 21 days. Rabbits can be kept for longer period of time for continued immunization and production bleed. Rabbit RPM provides speed, convenience, & flexibility to your polyclonal project.
