*What are humanized mice? *

Humanized mice express human proteins, allowing in vivo studies on human conditions. In some cases it is possible to replace an entire mouse gene with its human counterpart. Sometimes it may be necessary to keep some mouse sequences to ensure that the humanized protein functions properly in the mouse.

How are humanized mice generated?

A human cDNA or human genomic DNA is homologously recombined into the mouse genome with the aim to replace the functional domain of the mouse gene with the human one. This leads to the expression of the human protein in place of the mouse protein. Each humanization project is based on your requirements and we will assess your gene of interest to find the most appropriate approach.

*How does research benefit from humanized mice? *

Humanized mice aim to recapitulate the in vivo function of human proteins. Therefore, they are widely used to study and develop treatments for human diseases. Humanized mice are particularly helpful for pre-clinical drug validation of human therapeutic antibodies. In recent years, they have become invaluable tools for cancer research as models for immune checkpoint target validation and immuno-oncology.

What are knockout mice?

Knockout mice have been genetically modified to delete or inactivate a specific gene. The loss of gene function may cause a change in the mouse phenotype. When comparing human and mouse genomes, 80% of the genes have corresponding counterparts. Therefore, observing the characteristics of knockout phenotype can help researchers understand human gene function and genetic diseases.

How does Ozgene create conditional knockout mice?

Ozgene uses a conditional approach utilising Cre/loxP technology to generate your knockout model. The Cre recombinase excises DNA between two loxP sites. The loxP sites can be inserted around an essential exon in a gene. The mice are phenotypically wildtype but contain the floxed allele in all tissues. When bred to different Cre mice, tissue specific or inducible knockout mice are generated.

We continue to rely on the well-published technique of gene targeting via homologous recombination while monitoring and evaluating the reliability of the newer technologies for genome editing, such as CRISPR.

What type of research are knockout mice appropriate for?

Knockout mice are a valuable scientific method to study gene function. Conditional models enable the study of gene inactivation in a specific cell type, tissue of interest, or at a defined developmental stage. Knockout models are widely used for studying and developing treatments for human diseases, such as cancer, heart disease, diabetes, obesity, arthritis and neurodegenerative diseases. What are knock-in mice?

Knock-in mice have an insertion in a specific locus in order to produce genetically modified mice for customized research purposes. Reporter genes, human genes, related genes from the same organism, or point mutations can be introduced into the protein-coding region of a gene to produce knock-in mice.

What is Ozgene's method for producing knock-in mice?

We continue to rely on the well-published technique of gene targeting via homologous recombination to generate knock-in mice. The targeted modification is manipulated in embryonic stem (ES) cells in vitro. These manipulated ES cells are then used to generate knock-in mice with the desired modification. We recommend using a conditional knock-in approach (mini cDNA, Cre/lox inversion) to avoid deleterious outcomes that are often associated with non-conditional (constitutive) knock-in methods. This ensures controllable and inducible results with your knock-in mice.

Why generate conditional knock-in mice?

Our conditional knock-in mouse service eliminates risk that is often prevalent when a non-conditional (constitutive) knock-in method is used. Non-conditional knock-in mutations can cause ES cell lethality, chimera lethality and heterozygote lethality. They may also result in major disease phenotypes that impact on the breeding and maintenance of the newly created mouse strain. That is why we choose to create our knock-in mouse lines conditionally and germline transmission has been achieved for all projects to date. Our publications showcase successful research using Ozgene generated conditional knock-in mice. You can trust that we will strive to achieve the same outcome for you.

How are knock-in mice used in research?

Knock-in mouse models have several applications in medical research, especially to study and develop treatments for human diseases. Our targeted transgenic mouse projects now use goGermline, the revolutionary new technology to generate transgenic mice fast and efficiently. The aim of our targeted transgenic service is to generate a single mouse line, where the only genetic modification is the change in the targeted locus. This is not true for random transgenics, where you end up with multiple independent lines.

What do we mean by targeted transgenic mice?

Rather than randomly integrating the transgene into the mouse genome, the targeted transgenic mouse approach uses gene targeting by means of homologous recombination in ES cells. The transgene cDNA is knocked into a “safe haven” locus in the mouse genome to generate transgenic mice with a predictable expression pattern.

What is our specific method for producing transgenic mice?

Ozgene utilises the mouse gene, Gt(ROSA)26Sor, to generate targeted transgenic mice. The genomic structure of this gene, commonly referred to as ROSA26, is conserved between mice and humans. We use this technology since it provides you with the following benefits:

  • Only 1x transgenic mouse strain is needed. Not multiple founder lines that may or may not express your gene of interest.
  • This saves time, money, resources, genotyping, phenotyping, and animals.
  • The 3Rs (refine, reduce, replace) will be achieved for the Institutional Animal Care and Use Committee (IACUC).
  • Targeting into the ROSA26 locus achieves an ubiquitously inducible expression pattern.
  • Data is available for most tissues in the form of ROSA lacZ/eGFP reporter mice that have been crossed with many different tissue specific Cre mice.
  • A predictable transgene expressing animal is produced.
  • The ROSA targeted strain has more utility, as it can be used for tissue specific transgene expression just by using a different Cre mouse.
  • Unwanted side effects caused by insertional mutagenesis associated with random transgenic mice are avoided.
  • The targeted transgenic mice are readily bred to homozygozity.
  • You can directly compare data between targeted transgenic mouse lines. For example, various different point mutations can be compared in independent transgenic mice that you made or have been published.

Phenotypic characterisation of customised mouse models is important to your research. That is why we offer fully customised phenotyping that provides you with fast and convenient results. We provide validation phenotyping to verify the genetic change, as well as, exploratory phenotyping to further characterise your mouse model.

Some of our phenotyping services are listed below. However, as our service is fully customised, please contact us to discuss your specific phenotyping requirements and we will design a service to suit your needs.

What is validation phenotyping?

Validation phenotyping aims to verify that the genetic change on the DNA level in the mouse has also taken place on mRNA and protein levels, and the gene is behaving as we would expect. For example, it can confirm that the introduced gene is now expressed in the mouse, or that the gene that was knocked out is no longer having an effect on the model on the protein level. The presence of a genotype does not guarantee phenotypic expression.

What is exploratory phenotyping?

In addition to the validation phenotyping, we also offer customised phenotyping with individually designed experiments. This can include immune viability studies (such as in homozygous lethal models), basic morphological and/or behavioural analysis, immune checkpoint target validation, early determination of utility, characterisation under challenge, and other pre-clinical mouse research.

What phenotyping techniques are used?

Phenotyping can be performed on the mRNA, protein and in vivo levels, each subsequent level giving you more definitive information. Here are some of the phenotyping techniques used on each level:

  • mRNA expression: qPCR
  • Protein expression: IHC, ELISA, FACS, confocal
  • in vivo: Immunisation

Which tissues are analysed?

Confirming expression on the mRNA and/or protein level requires analysis of specific tissues. The tissues required are unique to your project. We are experienced in advising on the most appropriate tissues for your model in order to provide you with the most accurate results.

Once your new mouse line has been generated, you can utilise the convenient additional breeding options here at Ozgene. Our Vivarium provides the perfect site for your mouse breeding needs. It is Specific Pathogen Free (SPF) and complies with all major national and international research institutes.

We use our Geneoz VMS web-based management platform to manage all mice at our facility, ensuring effective and efficient breeding services for your project.

All breeding is customised to your project needs. Please see a list of additional breeding services below or contact us for a customised breeding plan.

  • Cre breeding: Cre knockin mice are bred to your gene targeted mice. The Cre gene is expressed early in embryonic development resulting in the removal of floxed sequences. We can also breed your mice to remove the Cre gene. Your gene targeted mice that carry the Cre gene will be mated with wild-type mice of the appropriate genetic background. Gene targeted offspring lacking the Cre gene will be determined by Southern Blot Analysis.
  • Flp breeding: Flp mice are bred to your gene targeted mice. The Flp gene is expressed early in embryonic development resulting in the removal of sequences flanked by FRT sites. It is also possible to remove the Flp gene from the genome of your gene targeted mice. Your gene targeted mice that carry the Flp gene will be mated with wild-type mice of the appropriate genetic background. Gene targeted offspring lacking the Flp gene will be determined by Southern Blot Analysis.
  • Tissue specific breeding: Many tissue specific Cre strains have been published and are available from various sources, most commonly The Jackson Laboratory. They can be imported on your behalf for mating to your gene targeted mice. Gene targeted offspring carrying the tissue specific Cre gene will be determined by Southern Blot Analysis.
  • Line maintenance: We create a small breeding colony from your mice of choice. This service provides you with a backup and therefore, peace of mind whilst your mice are being shipped or you are establishing a breeding colony in your own facility. This ensures that breeding age mice are available to you at any time.
  • Homozygous lines: This service is recommended in cases where heterozygous gene targeted modification does not result in an obvious or observable phenotype due to the presence of the remaining wild-type allele. We will breed your heterozygous gene targeted male and female mice to generate offspring homozygous for the gene targeted modification. Offspring are then genotyped by Southern Blot Analysis. You can expect that 25% of offspring to be homozygous, 50% to be heterozygous and 25% to be wild-type.
  • Colony Production: We can establish and maintain a colony at a predetermined size, age and gender, made available for regular shipments. This production colony will be designed to specifically suit your requirements for one or more colonies.
  • Custom breeding: Custom breeding is exactly that, customised to your needs. It could include, for example, transferring your gene targeted locus onto another strain background over several generations. This service is usually discussed on a project by project basis.
