Oxford Genetics mission is the creation of transformative systems and services to aid in the discovery and development of biologics, cell and gene therapies. A unifying theme across the portfolio is expertise in designing DNA and using this to optimise the expression of proteins, improving viral delivery systems and cell line engineering. Founded in 2011, Oxford Genetics is headquartered in Oxford, UK. Its products are sold directly and through a global network of distributors.

Synthetic Biology is a rapidly emerging and exciting new discipline that relies on complex genetic engineering strategies to build new biological systems that push the boundaries of genetic research. Oxford Genetics’ custom CRISPR singles, arrayed libraries or pooled libraries comprise a Cas9 expression plasmid, each containing a guide RNA (gRNA) toward a different target gene. These can be delivered into cells using standard transfection reagents, enabling rapid, effective knockout of target genes, and making them suitable for typical screening workflows. The singles and libraries are constructed on demand using gRNA designs produced by our proprietary design algorithm, which incorporates the latest developments in research to select the optimal gRNA designs, or you can supply your own gRNA designs and we can take care of the rest.

AAV Design

AAV vectors are created by replacing the Rep and Cap genes with the therapeutic gene, which is flanked by long terminal repeats (LTRs). The Rep and Cap genes are instead provided on a second plasmid, which is important as this provides all the viral genes that are essential for capsid formation and virion maturation. LTRs play an essential role in integrating the therapeutic gene into the genome of the target cell. Therefore, AAV vectors usually comprise of two plasmid expression systems.

Lentivirus Vector Design and Cloning

Lentivirus vectors are created by replacing the Gag, Pol and Env genes with the therapeutic gene, which is flanked by long terminal repeats (LTRs) and the psi-sequence of the retrovirus. The Gag, Pol and Env genes are instead provided by either one (2nd Generation) or two (3rd Generation) packaging plasmid(s) or a packaging cell line. Packaging plasmids or cell lines are important to produce viral vectors; these basically provide all the genes or viral proteins that are essential for capsid formation and virion maturation. LTRs play an essential role in integrating the therapeutic gene into the genome of the target cell, whereas the psi-sequence is essential for packaging the viral RNA along with the therapeutic gene into a virion. However, a single recombination event between the therapeutic genome (psi+) and the retroviral helper genome (psi-) can result in production of a replication competent wild-type virus. New packaging lines in which the lentiviral helper genome is split to prevent recombination have been developed. Therefore, lentitvirus vectors usually comprise of three or four plasmid expression systems.

Adenovirus Design and Cloning

Adenovirus vectors can be generated by insertion or deletion of DNA in three regions, namely the E1 gene, E3 gene and a short segment between the E4 gene and the end of the viral genome. First generation adenoviral vectors were developed by replacing the gene sequence at the E1 region with the therapeutic gene. Deletion of the E1 gene inhibits viral replication inside host cells. However, the transcription of the remaining viral genes resulted in early innate host cytokine transcription and adverse immune reactions. Thus, expression of the therapeutic gene was transient due to the immunogenic reaction that resulted in the destruction of all transduced cells.
Second and third generation adenovirus vectors were developed by deleting E1, E2 and E4 genes. The deletion of the above mentioned genes resulted in a reduced immune response against the vectors. Moreover, these vectors were also less immunogenic and showed long-term gene expression within the host. As mentioned earlier, the E3 gene can also be deleted in order to accommodate large transgenes.
