The facility was established in 2007 to assist Australian biotechnology companies and academic researchers to bridge the gap between laboratory experiments and the well-characterised cell line and bioprocesses required to produce material at the pilot scale. The NBF has a proven track record in mammalian cell line development and recombinant protein expression and purification at the Queensland node, and various expression systems at the CSIRO's Victoria node.

The Australian Institute for Bioengineering and Nanotechnology (AIBN) at The University of Queensland (UQ) houses the Queensland facility in a specially designed $13 million facility of labs, clean rooms and state-of-the-art equipment. The NBF has a staff of scientists and bioprocess engineers with world-class expertise in molecular biology, antibody engineering, mammalian cell culture, biopharmaceutical development and associated current Good Manufacturing Practices (cGMP).

Specifically, the facility services external clients in the areas of antibody discovery and engineering, mammalian cell line generation, upstream production, downstream processing development as well as protein analytics.

Antibody engineering:

  • Antibody discovery service using high quality phage display libraries of human antibodies. Antibodies can be isolated against a diverse range of antigen targets, including purified proteins, peptides, nucleic acids, or against cell-bound receptor antigens.
  • Construction and panning of both naïve and immunised phage libraries.
  • Antibody engineering services include cloning of hybridoma antibodies, production of bispecific antibodies, antibody fragments and antibody fusions.

Cell line generation:

  • Mammalian expression platforms designed for the long term expression of recombinant proteins such as monoclonal antibodies. Commonly used mammalian systems include Chinese Hamster Ovary (CHO), Human Embryonic Kidney 293 (HEK293) and hybridoma cell lines.
  • The facility also harbours a number of high throughput tools for cell line generation. These include;
    • ClonePix FL for directed semi-solid isolation of high expressing clones.
    • FACsARIA III for cell line enrichment and clonal isolation.
    • Clone Select Imager

Upstream processing:

  • Research to pilot scale bioreactors for batch and continuous culture.
    • Multi-well plate, standard shake flasks with Kuhner and INFORS-HT shakers
      incubators for research scale.
    • 1L/2L bioreactors for bioprocess development and DoE studies.
    • 2L/5L with Alternating Tangential Flow (ATF) technology for perfusion based studies.
    • Multiple WAVE reactors for standard fed-batch processes.
    • 50L Single-Use Bioreactor for pilot scale and clinical relevant products.
    • Collection of real-time tools for the measurement of cell density as well as gases and
      metabolites. For example; Cedex HiRes, Bioprofile Flex and capitance based online
      cell density sensors.

Downstream processing:

  • Large scale buffer preparation with the single use Mobius Mix 10-50 system.
  • Bulk harvest and clarification through depth filtration.
  • Protein A/L based affinity, IMAC and Ion-exchange chromatography with the AKTA Pure,
    Advant and Explorer systems.
  • Buffer exchange by cross flow filtration with the Cogent M1 Tangential Flow Filtration (TFF)
    and Pellicon stainless steel systems.

Phage display is a powerful method for discovery of new antibodies against specific target antigens. Libraries can contain over 1 billion different antibody clones which are displayed on the surface of bacteriophage and are screened against a desired antigen. Libraries can be either naive (where the antibody sequences are cloned from an individual or population without prior exposure to the antigen) or can be immune (where the sequences are cloned from an immunised animal or a patient with a particular disease or antigen exposure). National Biologics Facility has experience in creating both naive and immune human libraries, and immune mouse libraries.

For antibody discovery, the library is panned against the purified protein antigen or peptide, or cell-surface antigen, to isolate specific clones with the desired binding specificity.
