CAMELID SINGLE DOMAIN ANTIBODY PLATFORM
NanoGetic team is composed by scientists with recognized experience in the generation of camelid single-domain antibody (sdAb) phage display libraries. Camelids produce a fraction of a unique G-like immunoglobulin isotype that is naturally devoid of the Light chains, named Heavy-chain antibodies (HcAb). The variable domains (VHH) of these HcAbs can be obtained, through recombinant gene technology, as sdAbs. These recombinant sdAbs are the smallest available intact antigen-binding fragment (12-15 kDa) harboring the full antigen recognition capacity. Their exceptional physicochemical properties, possibility of humanization and unique antigen recognition properties make them excellent candidates for targeted delivery of biologically active components.
Immune libraries. NanoGetic S.L. construct these libraries by cloning the VHH genes repertoire, produced by peripheral blood lymphocytes of immunized dromedary with the antigen of interest, in a property phage-display plasmid to finally select antigen-specific VHH fragments (sdAbs).
Non-immune (naïve) libraries. NanoGetic own a larger dromedary sdAbs phage display library to select naturally occur specific sdAbs.
Humanization. sdAbs can be humanized by changing the amino acids 49 (E–G) and 50 (R/C–L) in the framework-2 maintaining their properties and specificity.
Tailoring sdAbs by genetic engineering to generate: o Multivalent immune tools: two or more linked sdAbs (bivalent, trivalent, etc.) o Multispecific immune tools: linked sdAbs which recognized different antigens (bispecific, trispecific, etc.).
