Polyclonal antibody development can be seamlessly integrated into a project when the goal of having a matched pair is defined, or if you require ADA assay reagents alongside your anti-idiotype monoclonal development. MBS offers rabbit, sheep, and goat polyclonal antibody development. A collection of production bleeds begins upon consultation with the customer when the screening data indicates that the animal has generated sufficient titer and specificity for the end application. MBS offers purification of polyclonal sera by protein A, protein G, and antigen-specific affinity purification and Ig polishing. Based on years of purification experience with a vast number of cell culture supernatant, ascites and serum samples, our purification team has developed optimized protocols to generate high quality antibody in quantities sufficient for early screening studies through large-scale production.

MBS offers both in vivo and in vitro antibody production of monoclonal antibodies in our cGMP facility. Antibody productions through mouse ascites provides customers with a cost-effective option for large scale antibody productions. In vitro production methods are available when smaller quantities of antibody are needed or a preparation free of endogenous mouse antibodies is required and the option to produce in serum free media is preferred. Production schedules are customizable and projects can typically be started within two weeks of booking.

In vitro Monoclonal Antibody Productions via Roller Bottle (3-4 weeks)

  • All incoming cell lines require MBS mycoplasma testing. Discuss mycoplasma decontamination services for lines that may be contaminated.
  • Cultures are grown in custom media and harvested for optimal antibody yield

Determination of an antibody’s unique characteristics is essential for the development of a successful assay. Customers may also consider antibody characterization services to confirm lot to lot consistency. Antibody characterization services available at MBS include:

  • Enzyme-Linked Immunosorbent Assay (ELISA): Confirm antibody specificity to the customer supplied antigen in an immunoassay format.
  • Western Blot: Confirm antibody specificity to the customer supplied antigen in an immunoblot format.
  • Size Exclusion Chromatography (SEC): MBS offers purity analysis of antibody samples by size exclusion chromatography. SEC separates monomeric IgG from dimeric, aggregated, and fragmented antibody under native conditions. A percent purity is calculated by peak integration and a report is provided.
  • SDS-PAGE: SDS-PAGE evaluates purified antibody samples under non-reducing and reducing denatured conditions. This electrophoretic characterization identifies intact IgG as well as heavy and light chain components.
  • Isoelectric Focusing (IEF): IEF analysis is run on purified monoclonal antibody samples to determine the isoelectric point (pI). This is the pH at which the antibody carries no net electrical charge and can be useful in determining suitable buffer conditions. The electrophoresis technique can resolve an isoform profile which can be used for evaluating lot to lot consistency.
  • Endotoxin Testing: MBS offers quantitative LAL endotoxin testing of samples to determine the EU/mg bioburden. This service is offered using the Endosafe®-PTS system.

MBS has long used Protein A with pH gradient elution for the purification of murine monoclonal IgG antibodies. By providing the gentlest conditions possible we have been able to offer our customers consistent monomer IgG purity of ~95%. When circumstances warrant, we also offer the highest quality standards in Protein G and Immunoaffinity Chromatography techniques. Increasingly, however, our custom antibody development customers have therapeutic research goals that may require an even higher level of antibody purity.

To meet those customer’s needs, MBS is now offering an additional Size Exclusion Chromatography polishing step to be added to one of our standard purification protocols. Removal of misfolded and confounding IgG dimers and higher molecular weight aggregates consistently results in antibody preparations >99% pure. Soluble aggregates can cause non-specific background problems, loss of sensitivity and variability in immunoassays that can severely confound clinical studies.

Western blotting is an immunoblotting technique used for the detection of proteins that have been separated by SDS-PAGE and transferred to a solid membrane support (typically nitrocellulose or PVDF). Not all antibodies work well in a western blot assay, due to the fact that the protein is denatured and the epitope (antibody binding site) of the protein of interest may not be accessible. Specific screening may be required to detect antibodies that are compatible with western blotting and optimization of the assay conditions to maximize their performance.

In an effort to save time and money, MBS screens potential candidates as early in the process as possible to determine their utility as a western blot reagent. This initial screening helps to identify the best candidates to scale up for use in western blotting assays.

Following the identification of specific antibody clones, optimization of the conditions for western blotting is typically necessary. While the screening is typically done with a purified protein, assay development would be performed with appropriate cell lysates or tissue homogenates as appropriate.

At the most critical juncture in a successful antibody development project, MBS enables our customers to efficiently and affordably select the most suitable clone for their application. By providing purified antibody earlier in the hybridoma development process, a MultiPure clone library allows for an unprecedented opportunity to generate data including specificity, relative affinities, matched pairs and more.

Advantages

Rapidly generates purified antibody from up to 94 fusion product supernatants at a time

Technology

Fusion products are expanded and supernatant is collected in up to 15mL quantities. The antibody is bound to a resin and eluted into a sealed 96 well plate. The typical yield for the process is 250ug-1mg per well. The range of concentrations seen with the MultiPure process is 0.3mg/mL-1.0mg/mL. Once purified, the antibody can be biotinylated and/or sterile filtered. Customers may also choose to add sodium azide or request custom buffers.
