The In-Cell Western (ICW) Assay is a quantitative immunofluorescence assay performed in microplates (96- or 384-well format). It combines the specificity of Western blotting with the reproducibility and throughput of ELISA. In-Cell Western™ assays are also called cytoblots, cell-based ELISA, In-Cell ELISA (ICE), and FACE (Fast Activated Cell-based ELISA). With In-Cell Western™ assays you can:

Detect proteins in fixed cultured cells using target-specific primary antibodies and IRDye® fluorescent secondary antibodies
Quantify two targets at 700 nm and 800 nm, using spectrally-distinct infrared dyes
Quickly, accurately measure relative protein levels in many samples
Detect proteins in situ in a relevant cellular context

LI-COR offers three levels of Western blot service: 1) Imaging Only; 2) Advanced Imaging (Reagents and Near-infrared Imaging); and 3) Full-Service (Processing and Chemiluminescent or Near-infrared Fluorescent Imaging).

Your project will begin with a discussion of requirements, reagents, and detection methods. Both chemiluminescent and near-infrared fluorescent detection methods are available
Imaging Only Service is designed for researchers who currently have the necessary reagents and time for preparing a Western blot, but do not have the means to image the blot.

For chemiluminescent imaging:
• Submit a dry blot to LI-COR
• LI-COR incubates with an HRP-conjugated secondary antibody
• LI-COR applies a high sensitivity substrate
• LI-COR images on the Odyssey® Fc Imager

For near-infrared fluorescent imaging:
• Prepare your blot on nitrocellulose or low fluorescence PVDF (i.e. Immobilon®-FL) membranes*
• Incubate with IRDye® secondary antibodies, or antibodies labeled with fluorophores in the 700 nm to 800 nm range
• Submit a dry blot to LI-COR
• LI-COR images on the Odyssey CLx Infrared Imaging System

Your completed project will include image files for analysis in the following formats: jpeg, tiff, and Image Studio™ files. Image Studio files can be viewed and analyzed using Image Studio Lite (a free download), or the full version of Image Studio Software.

Near-infrared fluorescent imaging
• 1-10 blots** $350. Each blot after 10 is an additional $25 / blot
Chemiluminescent imaging:
• 1-10 blots** $450. Each blot after 10 is an additional $45 / blot
*These membranes are preferred for their reduced background when imaging in the near-infrared wavelengths.
**Minimum order of 10 blots per imaging method

Advanced Imaging Service is designed for researchers who are interested in performing near-infrared Western blots, but lack the necessary reagents or instrumentation. LI-COR supports near-infrared imaging in the 700 nm to 800 nm range, and recommends the use of IRDye® secondary antibodies for this type of processing. The reagent kit provided will include membrane, blocking buffer, and IRDye® secondary antibodies.

For processing and imaging:
• LI-COR sends you optimal reagents for ten near-infrared fluorescent Western blots
• Prepare your blots using these reagents
• Submit dry blots to LI-COR
• LI-COR images the blots on the Odyssey® CLx Infrared Imager

When your project is complete you will receive images for analysis in the following formats: jpeg, tiff, and Image Studio™ files. Image Studio files can be viewed and analyzed using Image Studio Lite (a free download), or the full version of Image Studio Software.

Full-Service Western blotting is designed for researchers who are interested in performing Western blots but lack the necessary reagents, instrumentation or time. The service is also useful if you need to troubleshoot. LI-COR scientists will perform the entire Western blotting procedure from start to finish.

For full-service Western blotting:
• Send your sample and primary antibody to LI-COR
• LI-COR generates an optimized protocol
• LI-COR performs the experiments

When your project is complete you will receive an optimized protocol and image files for analysis in the following formats: jpeg, tiff, and Image Studio™ files. Image Studio files can be viewed and analyzed using Image Studio Lite (a free download) or the full version of Image Studio Software.

Additional fluorescent detection channels and resolutions are available using an Olympus IX81 microscope.
