Combinatorial Peptide Libraries

In order to find the best active peptide out of all possible peptide sequences it is necessary to screen as much as possible of those sequences. A combinatorial library is able to meet that requirement. It is the systematic synthesis of a set of peptides. The synthesis and screening usually goes over several stages in order to find the best fit out of all possible peptide sequences. Although we focus on the synthesis of the peptide libraries, under certain conditions we are also able to screen them. The price and processing time depend on the size of the library and the length of the peptides.

To permit our clients to fully benefit from our unique proteomics services without the hassles of cell culture, treatment and harvesting, subcellular fractionation, protein assay and shipping of frozen lysates, we are pleased to offer the convenience and cost savings of our In Vivo services. Clients can select from over two hundred pre-made cell and tissue lysates at no extra cost for use in our Kinex™ KSAM antibody microarray or Kinetworks™ KCSS immunoblotting services. The cell lysates were prepared from our panel of 12 human tumour cell lines (described below) that have been incubated with diverse drugs and hormones in dose response and time course studies. The organs and tissues have been harvested from healthy monkeys, mice, rats and frogs.

We can also produce lysates from our human tumour cell lines prepared according to our clients’ requirements.

Send us your experimental compounds (e.g. drugs), proteins (e.g. cytokines and hormones) or oligonucleotides (e.g. RNAsi), and we will perform the treatments of the cells according to your specifications and generate lysates compatible for testing with any of our Kinex™ antibody microarray and Kinetworks™ immunoblotting services.

For use in our In Vivo services, we have selected 12 of the most commonly studied human tumour cells, which also provide broad representation for tissue source, gender and age of the originators. These cells were obtained at low passage number from the American Type Culture Collection. They have also received extensive previous characterization by Kinetworks™ analysis, and this data is available via the Internet from our KiNET databank.

But it is possible to release the peptides from the membrane as kind of small-scale peptide synthesis, usually done on large spots. The price depends on the number of peptides, their length and possible modifications.

Description of technique: Synthesis of peptide arrays onto cellulose membranes. The peptide spots have a diameter of about 2 mm or 7 mm.

Please provide the following if possible when requesting service:

  • How many peptides? The more peptides the higher is the price

  • How long are the peptides? The longer the peptides the higher is the price

  • What for type of membrane is requested? We offer different types of cellulose membranes in the price range of 20 to 200 USD.

  • What for spot size is demanded? The large spots cost 50% more than the small ones. The large spots are usually useful only if the peptides have to be released from the cellulose e.g. for analytical purposes.

  • What for modification of the peptide is requested? Each modification comes with a distinct price.

Small-scale peptide synthesis and subsequent transfer onto glass slides. Usually, the peptides will be spotted as duplicates or triplicates. This service is recommended if large number of copies of the same array has to be probed. The price depends on the number and the length of the peptides as well as the number of arrays and glass slides. In order to provide each customer with fresh high-quality arrays, we sell the slides only in batches of 20.

Description of technique: Synthesis of free peptides and transfer onto glass slides as microarrays. The peptides will be spotted in duplicated or triplicates.

Platform(s) used in the above technique: Solid-phase peptide synthesis; spotting onto glass slides.

Please provide the following if possible when requesting this service:

  • How many peptides? The more peptides the higher is the price

  • How long are the peptides? The longer the peptides the higher is the price. Minimum charge is for 8 amino acids even if the peptide is shorter.

  • What for purity is requested (crude, 80%, 90%, 95%)? The higher the purity the higher is the price. Recommended is the crude form for screening purposes.

  • What for modification of the peptide is requested? Each modification comes with a distinct price. Important for the attachment to the glass surface is that the peptides contain either free amino group(s) outside the active region (or additionally attached to the peptide) or an additional free cysteine.

  • How many arrays? The more arrays the higher is the price. In order to give every customer the best (freshest) quality of arrays we have to sell the slides in batches of 20 as we are getting it. That means that the minimum order of arrays would be also 20 (1 copy per slide, but it could be up to 80 (4 copies per slide)

  • Should the C-terminus of the peptide be a free acid or an amide as it is in the natural peptide bond? We recommend the amide. It is our and also some customer’s experience that the acid sometimes interferes with interactions unless the very C-terminus of a protein or fragments from digestion are investigated. The choice of the type of C-terminus does not affect the price.

  • What type of modification of the peptide is requested? Each modification comes with a distinct price.

Kinexus offers standard and custom Western blotting services for monitoring changes in cell signaling proteins. There are 4 standard phosphosite screens which track between 38-44 phoshosites based on themes of broad signaling pathways, cell cycle status, protein kinases and substrates of kinases; and 2 standard screens for measuring the expression levels of 75 protein kinases and 29 protein phosphatases. For our custom services, clients can choose any 18 antibodies/sample from our listing of more than 850 antibodies (or their own) or choose from 1-3 antibodies of varying MW probed against up to 8 different lysates. Regular pricing for our services depends on whether the samples are considered confidential or non-confidential.

Sample requirements:
Clients can send their own lysates for analysis, or we can prepare lysates according to client’s specifications with tissues or cell pellets.
