High Throughput Protein Expression

Kemp Proteins has incorporated the use of IMCStips® with our Hamilton Microlab STAR to provide clients with a flexible, high-throughput, microvolume solution to deliver purified microquantities of their proteins. We can supply up to 500µg of purified protein from several hundred transient expressions to enable our client to compare, prior to selecting the protein(s) to move forward into further studies. Using our high-quality automation to produce small volumes of proteins that provide consistent results with high accuracy, precision, and repeatability to screen your candidate proteins increases overall efficiency of your development program.

 

Kemp Protein’s Specialty Purification Process
Client communication: We consider this vital to maximizing the potential for a successful project outcome. Our initial discussions are information gathering against a set of questions. The information gained enables us to generate a statement of work, a “project design”, that is shared with you. Together we refine the plan until you are in agreement that we have captured the required specifications and deliverables for the project together with and nuances that may be particular to your project. Throughout the development of the Stable Cell Line we will supply regular updates through conference calls or information download through electronic means such as “Box”.

 

Project Commencement: The project begins when we agree on the design specifications and you provide us any client specific deliverables. Each project is different but runs to a general set approach.

 

Project Approach: Our approach to specialty purification development is dependent on the project being undertaken. However, they all have certain commonalities. Using a recent project as an example we have outlined the steps for this project – other projects and resins have different steps. We have found this an efficient and effective process to achieve the desired outcome of our clients.

The process starts with the preparation of the test sample which may come from a transient or stable cell culture and may be mammalian, insect or bacterial. When the supernatant or cell pellet has been prepared ready for purification the sample is placed in an appropriate sized tube on the Hamilton STAR deck along with equilibration, wash and elution buffers together with the appropriate IMCStips.

Monoclonal Antibody (mAb) Production

Kemp Proteins has a long history of producing monoclonal antibodies—both IgG and IgM—from a variety of hybridoma cell lines, including mouse, rat, guinea pig, rabbit, and human. We deliver milligram to gram quantities of low-endotoxin and purified antibody to customer specifications, supported by quality documentation (including a Certificate of Analysis).

Kemp Proteins is flexible in production scale (mg to gm quantities) and, once we have assessed the hybridoma, we guarantee production output. When possible, we transfer hybridoma production into serum-free media to facilitate purification and low endotoxin levels. We offer several production platforms appropriate to client needs and quantity, including single-use bioreactors (including Wave), spinner flasks, stirred tank, membrane flasks, and roller bottles.

Hybridoma Source: We accept hybridoma cell lines directly from clients or from third-party sources, or we can develop hybridoma cell lines for the project. If the hybridoma cell line is sourced from the client or from a third party, we require:

Antibody Purification: Our purification group provides both fully automated (BioRad NGC & GE AKTAPilot) and manual purification techniques performed in an ISO 7 / Class 10,000 environment. Our team is experienced with most standard purification media, including Protein A, Protein G, Protein A/G, and LigaTrap. Final buffer requirements are specified by the client.

Baculovirus/Insect Cell Protein Expression

Kemp Proteins’ expertise in the baculovirus expression system (BEVS) is well known in the industry, and our team has completed thousands of baculovirus protein expression projects. Over nearly three decades, we have developed robust protocols for the creation of baculovirus constructs, the characterization of expression conditions, and scaleup of protein expression and subsequent purification.

In addition to generating standard baculovirus high-titer stocks, we routinely produce baculovirus-infected insect cells (BIIC) that are cryopreserved as cell banks for use in various expression protocols. We can use three cell lines for production: Sf9, Sf21, and T.ni. We also offer virus plaque purification services, master virus banks, and working virus bank production.

Kemp Proteins has also developed a proprietary PEI-mediated transient transfection protocol using Sf9 and S2 cells for protein expression in shake flasks and stirred tank bioreactors. One advantage of this approach is the ability to rapidly screen variant constructs prior to production. We have been very successful in using this technology to produce VLPs in Sf9 cells at the 2-liter bioreactor scale.

Using these approaches, we have delivered a range of customer-defined proteins, including many difficult-to-express, novel proteins (never previously expressed) that have required our scientists to be innovative in their approach. Baculovirus Titering Service

Price on request

Kemp Proteins determines the infectious titer of baculovirus particles in various types of samples using the standard plaque assay and/or the Virocyt Virus Counter. Plaque assay titers require a week for results to be finalized while titers are available using the Virus Counter in less than 24 hours from the receipt of your sample. By comparing results obtained with these two methods we have developed a numerical factor that converts the virus-particle count obtained with the Virocyt counter to plaque forming units. For the plaque assay, plates of Sf9 cells are exposed to dilutions of the virus sample and covered with an agarose layer. The plates are incubated for one week at 27ºC and stained with a neutral red dye. The titer of the virus is determined from the mean of two wells at two selected dilutions of virus. The results are presented as pfu/mL HTS. For the rapid Virocyt Virus Counter method samples are diluted and stained with a combination dye containing fluorescent tags for protein and nucleic acid. The counter detects particles that demonstrate coincident signals from the protein and nucleic acid probes above a certain threshold value. These coincident signals register as a virus particle. In addition to baculovirus this method has been used to successfully determine the titer of various other types of viruses including influenza.

Bacterial Protein Expression

If complex post-translation modifications are not required for proper folding and function, bacterial expression is an economical way to produce recombinant proteins.

Kemp Proteins provides protein expression and plasmid production services using numerous commercially available strains of E. coli cells. We produce recombinant proteins at scales that range from 50 mL to 10 liters using shake flasks and stirred-tank fermenters. We offer gene-to-protein services to produce HIV proteins, scFv, enzymes, and mitochondrial and channel proteins. Our team has developed stable isotope labeling protocols to provide labeled protein for NMR studies. We use affinity, ion-exchange, and size exclusion procedures for protein purification. Proteins are dialyzed into PBS or a buffer designated by the client.

Kemp Proteins offers plasmid production to support protein expression using mammalian transient transfection protocols. We produce and purify low-endotoxin, high-purity plasmid in quantities that support expression in the 1-100 liter range (1-100 mg plasmid). Each batch of purified plasmid is tested for endotoxin and examined on agarose gels for purity and supercoiled state.

Please note: To facilitate affinity purification and/or solubility, most antigens produced in bacteria are fused to a tag.
