For example, if it is obvious early in a stem cell analysis that the “stem cells” have abnormal karyotypes, the client has the option to cut the analysis short and pay a lower price.
At KaryoLogic, research into innovative new cytogenetic techniques is ongoing. Thus, we offer services not found elsewhere, such as the Premature Chromosome Condensation Assay for quiescent cells. Currently, in collaboration with partners in academia and private industry, we are investigating new techniques in RNA-DNA FISH.

WHAT WE OFFER:

Karyotyping of human, mouse, rat, horse, and other species; stem cell lines, cancer cell lines, tumor and primary cultures; adherent and suspension cultures. We accept live cells shipped to our Durham, North Carolina location. Cells on feeder layers or basement membranes are not a problem.

A variety of options for the number of metaphase spreads analyzed: Simple G-Band Analysis for 5 to 10 metaphase spreads analyzed, Standard G-Band Analysis for 20 metaphase spreads analyzed, or Complex G- Band Analysis for 25 or more metaphase spreads analyzed, or a custom service.

Detection of chromosomal aberrations of all types: translocations, trisomies, aneuploidy, deletions, inversions, duplications, rings, dicentrics, breaks, double minutes, and much more.

Excellent banding resolution for detection of subtle chromosomal aberrations

Karyotyping of quiescent cells, and/or cells with a low mitotic index (as in irradiated cells) by the Premature Chromosome Condensation Assay.

Fluorescence in situ hybridization of DNA probes to metaphase spreads and interphase nuclei.

Reports that include karyotype images, data summary, and interpretation of results. High resolution chromosome photos for publication, tables, figures, and extra images available upon request.

All work conducted "in the spirit of" GLP (Good Laboratory Practice) meaning that KaryoLogic has incorporated many of the quality processes and standards consistent with GLP, however no claim of full compliance or certification is made at this time.

OUR SPECIAL ATTRIBUTES:

Rigorously trained and highly experienced staff with many years of experience, particularly in human clinical cytogenetics and mouse cytogenetics.

We offer a briefer, less expensive analysis, our “Simple Analysis”, for answering quick questions immediately obvious from the chromosome spreads – an excellent option for when stem cells unexpectedly turn out to be clearly abnormal.

Simple G-band Analysis – 5 to 10 metaphase spreads analyzed:

  • Recommended for answering simple, straightforward questions, e.g., Are these cells male or female? What specie is this culture? Is the karyotype abnormal?
  • Fee covers receipt of live cells, brief culture, harvest, fixation, slidemaking, banding/staining of chromosomes, and microscope analysis of 5 to 10 metaphase spreads.
  • Report includes summary and one representative image.
  • At the request of the client, this analysis can be extended to become the more detailed Standard Analysis if something of interest is discovered.

Standard G- band Analysis – 20 metaphase spreads analyzed:

  • Recommended for gathering typical information on chromosomal constitutions such as overall counts, gender, trisomies, monosomies, deletions, insertions, translocations, duplications, dicentrics, isochromosomes, rings, breaks, double minutes, polyploidy, endoreduplication, etc., and any other aberrations found.
  • Intended for cell lines that are diploid, or near diploid.
  • Fee covers receipt of live cells, brief culture, harvest, fixation, slidemaking, banding/staining of chromosomes, and microscope analysis of 20 metaphase spreads.
  • Report includes a written summary and two representative karyotype images.

Complex Analysis – 25 metaphase spreads analyzed:

  • Recommended for gathering more detailed information on highly abnormal hyperdiploid karyotypes, chromosome by chromosome, with details of breakpoints in chromosomes of interest. As in the Standard Analysis, overall count, gender, trisomies, monosomies, deletions, insertions, translocations, duplications, dicentrics, isochromosomes, rings, breaks, double minutes, polyploidy, endoreduplication, etc. and any other aberrations will be scored.
  • Fee covers receipt of live cells, brief culture, harvest, fixation, slidemaking, banding/staining of chromosomes, and microscope analysis of 25 metaphase spreads.
  • Report includes a written summary, two representative karyotype images, images of significant breakpoint details, and/or tables summarizing aberrations.

Interphase Chromosome Analysis using Premature Chromosome Condensation– 20 spreads from interphase nuclei analyzed:

  • Recommended for analysis of cells with low mitotic index, or unable to proceed through the cell cycle. This technique utilizes drug-induced chromosome condensation, as opposed to arresting cells at metaphase.
  • Fee covers receipt of live cells, brief culture, harvest, fixation, slidemaking, banding/staining of chromosomes, and microscope analysis of 20 chromosome spreads.
  • Report includes a written summary and two karyotype images.

Sample requirements: For each adherent cell line, prepare two T25 flasks with cells seeded at different confluencies. Fill flasks to the top with media, tighten caps, and wrap with parafilm. For suspension cultures, send 10 ml of cell culture (minimum 500,000 cells) in a 15 ml tube and include a tube of 10 mls media. For all samples, triple package and ship at ambient temperature by overnight carrier. Turn around time 7 to 14 days.
