An antibody generation approach based on the selection of antigen specific recombinant antibody fragments from proprietary human or llama phage libraries or from custom build immune libraries. Our proprietary libraries have been made from human patient and naïve repertoires, as well as from naïve llama (VHH and scFv) repertoire. Custom immune libraries are prepared from spleen, lymph nodes, and bone marrow of immunized animals or humans. This approach captures the immune repertoire of immunized animals or patients and allows sampling of this repertoire by semi-automatic panning procedures, to rescue and identify unique antibodies with pre-specified characteristics. Antibody fragments can be expressed in bacteria or rapidly reformatted into any given Ig format, such as full size IgG of various isotypes, Fc mutants to modify effector functions, or chimera with Fc regions from different species.

Standard Method Monoclonal Antibody Development in Mice & Rats

Intraperitoneal injections and a choice of adjuvant is available to suit the clients specific needs. When this traditional immunization procedure is coupled with our Single-Step cloning method, weeks are shaved off the timeline of monoclonal development.
A novel immunization platform for the generation of monoclonal antibodies against challenging and difficult to express conformation-sensitive targets, including complex protein classes such as ion channels and GPCRs. Within a DNA immunization approach, the antigens will be expressed in vivo in the animal and thereby go through the post-transcriptional modification process and therefore the antigens maintain their naïve conformation. Freezing/Cryopreservation of Hybridomas (and other cell lines) Standard Operating Procedure
Most hybridomas are frozen in either growth media with 20% fetal bovine serum (FBS) and 10% dimethylsulfoxide (DMSO) or 90% FBS plus 10% DMSO and stored in liquid nitrogen. IPA freezes hybridomas in 90% FBS plus 10% DMSO in 1mL (2 x 10e6 cells/vial) aliquots. Always wash DMSO away when thawing. Thaw frozen vials directly from liquid nitrogen in a 37°C water bath for exactly 2 minutes. Immediately transfer cells to 10mL DMEM complete media plus 10% FBS in 15mL conical tube, mix gently by inverting several times, spin down at 200 x g for 6 minutes. Transfer pellet to 10mL DMEM complete media plus 20% FBS in pre-gassed 25cm2 flask. FBS content is reduced to 10% once cells are recovered (2-3 days post thaw).
Freeze in 1mL aliquots of 90% FBS and 10% DMSO at 2 x 10e6 cells per vial. Split cells ½ for at least 2 days before freezing while maintaining them at mid log. Freshly divided cells have stronger membranes that withstand the freezing process better. Keep freezing media ice cold during the freezing process as DMSO is toxic to cells at warmer temperatures.
