The High Resolution Plant Phenomics Centre (HRPPC) is the Canberra node of the Australian Plant Phenomics Facility and is located in Canberra at CSIRO Plant Industry and the Australian National University.

This centre focuses on “deep phenotyping” (delving into metabolism and physiological processes within the plant) and “reverse phenomics” (dissecting traits to discover their mechanistic basis).

Next generation research tools are applied to probe plant function and performance, under controlled conditions and in the field.

Recent advances in robotics, imaging and computing are used in applying these technologies and scaling them from the single plant to the ecosystem level. Two levels of service are provided in the HRPPC.

First, projects can be housed in the “Research Hotel” environment where screening systems can be developed using facility staff and resources then deployed in the facility and in the user's home institution.

Second, users’ material can be screened for specific attributes using one or more of the modules housed at CSIRO or the ANU. The HRPPC's focus is on flexibility from cereals to dicots and woody perennials at all stages of development.

The Crop Plant Field Module is an important part of validating controlled environment screening technologies and provides a powerful high throughput set of tools. The environmental conditions and plant performance in the field can be quantified with developing technologies like stereophotography, LIDAR, hyperspectral reflectance, leaf thermography, electrical root detection, sensor networks and other developing technologies. These technologies will be integrated into spatial, climatic and precision agricultural data bases by researchers across Australia. There is capability to validate GM plants in the field, while still adhering to regulatory requirements. GM plants were analysed last season at a field site and the study will continue this year. The Field Module utilises a number of technologies developed by the centre for field analysis such as The Phenomobile, Phenonet, Tethered Blimp and Phenotower.

This module is capable of conducting analysis into:

-Non-destructive estimate of biomass and crop structure pre and post-canopy closure
-Remote sensing of stress response, canopy water loss and photosynthetic response
-Remote sensing of chemical composition : CHO, protein N, pigments over entire lifecycle
-Application of distributed sensor networks for simultaneous continuous monitoring in the field (micromet plus low res versions of the above)
-Non-destructive detection of water and root biomass at depth in soil

APPF provides two approaches to field simulation;
(1) a semi-static facility, the Cropatron, with a high degree of environmental control, containment and high resolution monitoring and
(2) a mobile facility: the Field Mobile Cropatron which can be placed over part of a crop in the field to control the growth environment, or for a short term, the measurement environment.

In both cases a double membrane, inflatable transparent dome is placed over approximately 10m2 of crop under natural illumination.

In the static systems, the plastic dome can be moved on a gantry to enclose a plot for a short period or the entire lifecycle. Gas flow to and from the chambers and data cabling communicates through the gantry and services rail to the analysis laboratory containing infrared gas analysis (IRGA) and IT infrastructure for data capture.

This also allows flexible expansion of the facility. The Field Cropatrons will be mounted on a small cherry picker which can be towed with a 4WD vehicle.

We envisage demand can be accommodated with 10 static Cropatrons, which include two Pathatrons (pathogen containment suitable for disease challenge and monitoring experiments) and five mobile units.

A variety of levels of environmental control and monitoring can be provided.

Model Plant HTP Screening Module: Fluorogroscan
High Throughput Arabidopsis Screening :

-Growth and morphology
-Photosynthetic performance (Chl Fluor) under defined conditions
-IR screening for leaf temperature
-Automated destructive sampling for metabolites, protein, -DNA and RNA, 13C delta

Fluorogroscan was developed by Phytosphere Juelich. It uses a PAM imaging fluorometer to measure chlorophyll fluorescence and the image can be background subtracted to give projected leaf area for growth analysis and morphological characterisation.

The camera steps across trays of 20 plants using an X-Y-Z robot, illuminating with a flash of very high light then measuring chlorophyll fluorescence. False colour images area produced which represent calculations of chlorophyll fluorescence parameters on a pixel by pixel basis.

Regions of interest can be identified for screening in high throughput . Outputs can be false colour images for spatial information or CSV files of calculated values of dark adapted Fv/Fm (gives the “health” of the photosystems), commonly used in abiotic stress screening and for mutant screens (photoinhibition and pigment mutants).

Steady state Fm’ and electron transport rate in the light can also be calculated as a surrogate for photosynthesis.

This allows mathematical quantification of morphologies based on standard ontological plant descriptors. These can be graphed using polargrams or entered into a database (outputs CSV files) and PNG image files.

This has been tested to cluster phenotypes of ABA sensing mutants and works well. I will provide you with an example of analysis of heterosis in crosses of LER and C24 and inheritance of rosette architecture.
