HansaBioMed Life Sciences provides high expertise for purification of Large (>150 nm) and Small EVs (30-150 nm) from human or animal biofluids, cell conditioned media and plant extract. EV purification can be obtained following our standard protocols which combine Tangential Flow Filtration (TFF) and Size Exclusion Chromatography (SEC). Alternatively, differential ultracentrifugation and microfiltration, chemical precipitation or immunoaffinity methods can be used.
Isolated EVs are subsequently quantify for total protein contents and validated by nanoparticles tracking analysis (NTA) with Zetaview (Particle Metrix). Additional characterization can be performed by ELISA, FACS, TEM, Western blotting, on request. Purified EVs can be shipped to the customer lyophilized or frozen, as required.
Nanoparticle tracking analysis (NTA) is a method for visualizing and analyzing particles in liquids that relates the rate of Brownian motion to particle size. NTA allows the determination of a size distribution profile of small particles with a diameter of approximately 10-1000 nanometers (nm) in liquid suspension.
HansaBioMed Life Sciences performs NTA analysis using the Zetaview instrument of Particle Metrix, which allow EV characterization by size distribution, concentration, Zeta Potential. Additionally, the instrument allows EV phenotyping by NTA performed in fluorescent mode.
Extracellular Vesicle properties and marker composition depends on the state of parental cells/tissues. EVs shuttle proteins on their surface as well as into the vesicle. Together with RNA, proteins are an important source of biomarkers and potential indicators of disease states. HansaBioMed Life Sciences provides services for phenotyping the EVs by marker detection, quantification and profiling of membrane or internal proteins. Protein markers can be quantified via ELISA assay, flow cytometry, or fluorescence NTA analysis.
EVs shuttle functional RNAs (75% miRNA) and genomic DNA. HansaBioMed Life Sciences provides a wide portfolio of services for isolation of EV associated RNA and DNA from vesicles derived from human/ animal biofluids (plasma, serum, urine, etc.)and cell culture media. Additionally, our scientists can provide an expert service for identification of EV-associated RNA/DNA biomarkers leveraging the throughput and scalability of Ion Torrent and SOLiD System (Thermo Fisher Scientific) next-generation sequencing platforms. This service is an easy solution for researchers who are interested in identifying new EV RNA biomarkers or in the understanding the profiling expression of such biomarkers in the vesicles of their model cellular systems or patient biofluids.
Sequencing or profiling service takes approximately 4-6 weeks.
