No two research projects are exactly alike but it is important that the processes and procedures you use are predictable and replicable. Envigo has more than 20 years of experience refining our approach to developing monoclonal antibodies from a wide variety of peptides and proteins. While our procedures are well-defined, we are flexible when we need to be.

When you partner with us, our scientists will work with you to develop a procedure that will best meet your needs, with processes that are consistent and service that is responsive.

Our production process is broken down into 3 stages which allows you to pay for only those stages that are completed. Once the desired clones are developed, we will scale up some material for your in vivo or in vitro use.

Our schedule for producing up to 3 positive clones includes:

STAGE I: Immunization

Ten virus antibody-free mice will be immunized with the customer's antigen. The mice will be housed in an isolator during the immunization period. We immunize 10 mice to increase our chance of an immunogenic reaction. In order to get a better antibody response, the antigen is prepared with an adjuvant such as FCA (Freund's complete adjuvant) for the primary immunization. The emulsion contains heat-killed mycobacterium that ensures the antigen is released slowly into the animal's circulatory system. The bacteria in the adjuvant stimulates the animal’s immune system. Additional boosts or immunizations are necessary for high antibody levels. FIA (Freund's incomplete adjuvant) is used.

Common routes for injection are i.p., i.m., i.v., s.c. For hybridoma development, s.q. is typically used. 5-100 ug of antigen are used for each immunization. We ask the client for 2-3 mg of antigen.

At six weeks, following the primary immunization and the first boost, the mice will be tested for antibody titer. The antibody will be determined by ELISA assay unless the customer has an alternative assay. If additional boosts are required, they will be administered and the mice will be retested for antibody titer. Once immunization is complete, the mice will be held for three-to-four weeks before fusion. Three-to-five days before the fusion, a mouse will be selected for the pre-fusion boost.

STAGE II

  1. Fusion

The spleen from the mouse will be harvested and all spleen cells fused with a myeloma cell line. We generally use NS-1 for the fusion. Generally six-to-ten 96-well panels will be plated. Cells will be grown in HAT (Hypoxanthine, aminopterin and thymidine) selection medium. This selectively kills cells deficient in the enzyme (HGPRTase), which is essential for the operation of the salvage pathway for making nucleic acids. The myeloma and spleen cells will die off. A maximum of three separate fusions will be performed.

  1. Screen and Select

All 96 well panels will be screened by ELISA for antibody positive wells. The standard production will be screened with a single ELISA antigen. Additional ELISA antigens may be added as an option. About 15 positive wells from the ELISA assay will be selected to be expanded and frozen for possible cloning. Tissue culture supernatant (0.2 ml per clone) can be sent to the customer for further testing.

STAGE III

  1. First Cloning

A maximum of 6 positive wells will be selected for cloning. They will be cloned by the limiting dilution method. In this method cells are diluted so that they can be individually pipetted into separate wells. Isolated colonies will be tested by ELISA and positives selected to be expanded for the next cycle of cloning. Colonies should be visible after 10-14 days. Cells from these positive clones will be frozen as backup.

  1. Final Cloning

Positive clones from the first cloning cycle will be selected for the second cycle. Isolated colonies will be tested by ELISA and positives selected to expand. Several cycles of cloning may be required in order to obtain true, stable clones. Cells from these positive clones will be frozen as backup.

  1. Selection or Non-HT Requirement

A maximum of three positive clones will be further selected for clones, which grow in medium lacking HT.

  1. Frozen Stock and Ascites

The final three clones selected will be expanded and a small stock of each will be frozen. A small amount of tissue culture supernatant (2-5 ml) will be prepared for each clone. Each clone will be isotyped. In addition, several mice will be inoculated with each clone for ascites production. Supernatant production can also be performed. Ascites fluid, supernatant and/or vials of cells will be shipped to the customer for evaluation.

NOTE: Our service allows for the development of up to 3, 5 or 10 positive clones.

Envigo can fragment your antibodies to produce Fab and F(ab)2 fragmentation to your desired specifications. The fragments are then purified by Protein A and/or cation exchange chromatography. We also offer antibody and antigen conjugation services. Conjugation services available include:

  1. Biotin

Biotin is the conjugate of choice if the antibody is going to be used in a variety of applications. Biotin is a small molecule that has little effect on an antibody’s immunoreactivity. Its small size makes biotin an excellent tag for your antibody in most techniques. Biotin binds tightly to avidin or streptavidin, which are available enzyme conjugated, making your biotinylated antibody useful in most immunological procedures. The disadvantage of a biotinylated antibody over an enzyme or fluorescent conjugated antibody is that the biotinylated antibody involves an additional step in your assay.

  1. HRP and Alkaline Phosphatase

Enzyme conjugates give a highly selective and sensitive immuno-reagent. By directly labelling your primary antibody, you eliminate steps and washes. Horseradish Peroxidase is a stable enzyme suitable for ELISA, Western Blot and immunocytochemistry. HRP has low endogenous activity and a high turnover rate, producing its colored-end product in a short amount of time. Alkaline phosphatase is often used when high background is experienced with HRP interacting with endogenous peroxidases. Alkaline phosphatase allows linear reaction rates improving sensitivity by reacting over longer time periods.

  1. Fluorescent Markers

Fluorescent probes are used in multiple probe assays, immunofluorescence microscopy and flow cytometry.

Your research and manufacturing cell lines must be developed by experts who can be trusted to exercise the same degree of care that you do. As your partner, Envigo has exacting processes to meet your expectations.

Envigo production of monoclonal antibodies for use in your diagnostic or therapeutic applications is carried out in our cGMP, FDA-registered facility. Our service utilizes a multi-phase production process with an extensive quality assurance program under the guidance of our knowledgeable technical and scientific staff.

When you entrust your mAb production to Envigo, your project will receive the same consistent and reliable processes and procedures that your research and manufacturing environments require.

Receipt and Expansion
Upon receipt, all cell lines are assigned an Accession Number for tracking purposes. The cell line is transferred to a quarantine area where it is grown in culture and expanded to create a cell bank.

MAP Testing and Cell Storage
MAP testing is performed on the cell stock. Our standard MAP panel tests for 19 murine adventitious agents, including mycoplasma. Testing is completed in approximately six weeks. After MAP testing, the cell stock is stored in a cGMP, alarmed, cryogenic tank for future runs.

Optimization and Inoculation
Envigo offers aa unique approach to research optimization for large-scale production. This involves performing a dosage study to determine time frames for ascites production development, mortality, incidence of solid tumors, volume of ascites, and concentration of immunoglobulin.

Optimization saves time and allows maximum monoclonal antibody yield. The ascites can be used by the customer for analysis prior to large-scale production. As for your cost concerns? This unique service is provided at no additional cost for production runs of greater than 100 mice. Optimizations are available for smaller runs at an additional cost.

Production
With the MAP testing and optimization completed, production is initiated under cGMP guidelines. Complete line clearance is practiced at all points during the production process.

Harvest
Ascites is collected on wet ice and centrifuged to remove cellular and lipid material. The clarified ascites is placed into sterile containers, labeled and immediately frozen.

Quality Control
Quality control on the final product can include subclass-specific RID to determine immunoglobulin concentration, U.S.P. sterility testing, 9CFR sterility testing, and determination of specific activity by ELISA and endotoxin testing. Final ascites is stored frozen at either -25°C or -70°C until it is ready for shipment.

Ascites Production for Research Use
Ascites production for research includes the same phases as cGMP, however, MAP testing of the cell lines is not included thus reducing costs and decreasing turnaround time. Our technical staff will assist you at each phase of production. Should you need it, our protocols can be customized to meet your required specifications.

Are you concerned about the health status of your research models? We understand the impact of health status on the outcome of your experimental procedures. Your colony must be maintained within generally accepted target microbiological status specified within the FELASA Recommendations (FELASA, 2014). Envigo’s Rederivation service standardizes the reporting of microbiological status and allows animals to be exchanged between laboratories and breeders with the confidence that known microbial adventitious agents will not be inadvertently introduced.

When you utilize our Rederivation service, you can expect:

You want to preserve the integrity of your research colony’s genetic lineage, but you don’t have the capacity, time, resources or desire to maintain it until some undefined future date. That’s understandable, particularly as the number of genetically altered lines developed and maintained for research in animal facilities continually increases. Laboratories often develop their own strains that are genetically modified for specific purposes. The challenge is that your high value strains often result in continued maintenance even though you may not need them for long periods. Besides, such maintenance may produce excess animals and can be expensive, too. You can avoid these consequences. By utilizing Envigo’s Cryopreservation Service, you can:

Envigo is dedicated to providing the highest quality surgically modified animals. Surgical services personnel undergo rigorous training and evaluation prior to performing surgery, and surgical skills are continuously monitored.

Rodent surgical services are available from both North America and Europe. We will do our best to accommodate requests for overseas surgical procedures. Envigo also performs client-specific surgical procedures on rodents. For more information or pricing, please contact us to discuss your specific needs.

Envigo is dedicated to the humane care and use of research animals. All surgical modifications are reviewed and approved in advance by Envigo’s Institutional Animal Care and Use Committee (IACUC). Envigo’s veterinarians provide oversight and technical and professional support to surgical personnel.

All surgical procedures are performed by trained and tested surgeons using aseptic technique in surgical facilities dedicated to maximum-security production barriers or isolators. Surgically-modified models are monitored postoperatively to ensure complete recovery from surgery. All animals undergoing major survival surgery (as defined by the Guide for the Care and Use of Laboratory Animals, 2011) receive post-operative analgesia to further ensure their humane and ethical care. Exceptions to analgesic requirements will require pre-approval by Envigo’s IACUC.

Envigo recognizes the need to supplement enrichment to those animals needing to be singly housed after surgery (e.g., catheterized rats). Therefore, we provide these animals with a section of inert nylabone for gnawing or provide supplemental novel material for bedding. These enrichment items are provided to animals immediately post operatively and remain with them until delivery to the client.

The introduced 2010/63/EU Directive indicates that animal care staff shall be adequately educated and trained before they perform procedures on animals. Member States are obliged to publish the minimum requirements with regard to education and training and the requirements for obtaining, maintaining and demonstrating requisite competence.

When you work with laboratory animals, it’s your responsibility to have appropriate training and competencies as they ensure best practices and solid results in the best interests of both the animals and the science. To maintain competencies, current levels of skill and knowledge must be regularly assessed. Therefore Envigo is offering a range of training courses, which are listed below.

All courses are currently only available in Europe and are given by experienced trainees and external experts with a laboratory animal science background, coordinated by our Veterinary Science Group and Reproduction Specialists. They aim to develop knowledge and skills for scientists and technicians with a focus on best practice and scientific background. Courses have a strong focus on the individual participant, especially during the hands-on training sessions.

In addition to the range of hands-on surgical and reproductive training procedures, Envigo is able to support your staff with general laboratory animal courses. Our veterinary specialists share their extensive knowledge during ½ or full day seminars. Courses can be presented on-site or through webinars.

Laboratory Animal Courses

  • Rodent colony management, 1 to 2 days: Reproductive physiology, genetics and breeding schemes (colony management), nomenclature, euthanasia.

  • Breeding and genetics, 1 day: Breeding schedules, congenic, coisogenics and consomics. Genetically modified models (GEM’s).

  • Mouse and rat colony health care , 1/2 day: Common rodents diseases. Risk, treatment and management, quarantine, and rederivation.

  • Nutrition , 1/2 day: Fixed formula diets, research areas affected by presence of phytoestrogens. Diet should reduce rather than introduce variation.

  • Necropsy and sample collection (hands-on), 1 day: Full necropsies of mice and rats. Description of different analysis profiles. Terminal procedures, animal manipulation, necropsy techniques, blood and tissue collection, and bio-safety. The course does not require any basic surgical knowledge.

  • Introduction to cryopreservation, 1 day: Course starts with an introduction on cryobiology and then closely examines the use of cryoprotectants, freezing technologies, cryovials, straws and LN tanks. In addition, basic embryo and sperm freezing (Jax vs. Card methods) will be discussed.

  • Advanced course on cryopreservation and embryo transfer, 2 days: Two day advanced training course covering the basic principles of cryopreservation, embryo freezing and embryo transfer, as well as practical hands-on embryo collection, manipulation, freezing, thawing and culture.

  • Cryopreservation theory including cryoprotectants, 1 day: Course covers the theory and practice on sperm cryopreservation using the Jax and Card methods, including thawing, sample evaluation and theory on IVF. Slow and rapid embryo freezing including vitrification and freezing curves. Use of cryovials vs. straws. Mouse superovulation. Mouse euthanasia and embryo collection. Embryo culture. Mouse sperm freezing

Advanced course micro-surgery

Two day training course which builds on the introductory course and continues with basic information on anesthesia and peri-operative care of small laboratory rodents. The practical part will focus on more advanced techniques. This course has a very strong focus on the individual participant with a lot of hands-on surgery.

Telemetry in rodents

Two day training course designed to instruct participants on the applications of radio telemetry in small laboratory rodents both in demonstration and practical exercise. Basic skills of micro surgery required.

Invasive and/or semi-automated chronic blood sampling techniques in rodents

Recent developments including the availability of pre-catheterized rodents and high sensitivity bio-analytics have enabled chronic, serial blood sampling in rodents. The automation of blood sampling and the use of dried blood spot techniques have simplified the collection of micro samples, making it possible to take a full PK profile from a single animal. Surgery itself and catheter patency are critical to ensuring successful results.

This course focuses on surgical techniques of catheterization, acute sampling techniques (repeated) as well as the sampling methods including automated blood sampling with dried blood spot.

Injection and administration techniques in rodents

Designed to improve the competence of laboratory animal technicians who handle rodents for purposes of injection. Practical, hands-on training included. Many technical skills can only be obtained by intensive and careful training. We invite attendees to share knowledge and expertise.

Dedicated courses

We offer dedicated courses based on customer needs.

All courses are organized in the Netherlands, in dedicated rooms for theory and practice with a strong focus on animal welfare, since good welfare is good science. During all courses, we will provide information on practical items such as transport, housing and special care of surgically prepared animal models.

Technical Details

Science and medicine are focused on understanding and solving the diseases of aging. With an aging population globally, we know that demand for your research in this area is expanding. Envigo maintains a commercial inventory of male rat and female mouse models reserved monthly for aged animal availability, or can maintain cohorts of models upon request for your research needs.

Technical Details

Envigo provides ready availability of several rat and mouse models for your aged animal research. If the aged animals you need are not available from our inventory, or if the animals require three or more months of maintenance prior to shipment, Envigo will reserve and hold specific cohorts of animals until the requested age at shipment.

Aged animals are appropriate for use in the following types of research:

Female Mice:

  • C57BL/6JRccHsd (can only be sourced from Europe)

Other models for aging are available upon request, including C57BL/6NHsd mice.

Pricing:

Pricing and availability of aged animals from the aging inventory is available on request. For client-reserved aging cohorts, pricing is developed based on the requirements of the project. Prior to initialization of an aging project, Envigo will confirm your expectations, the project work scope, and animal pricing with an agreement of understanding.
