High-throughput sequencing of hybridoma variable regions using next-generation sequencing.

Features:

  • Full-length variable region sequencing of all clones

  • Only 40,000 cells per hybridoma needed!

We deliver:

  • Full-length variable regions for heavy chain and light chain for each hybridoma

You know the serum has the reactivity you need for you assay, so why look anywhere else for monoclonal antibodies?

Our proprietary Alicanto platform discovers natural, high affinity antibodies without creating a library. We use mass spectrometry-based analysis to deconvolve the polyclonal serum antibodies into monoclonals. 

Starting material:

  • 2.5 mg of antigen


Process:

  1. Animal(s) are immunized with client-provided antigen.
  2. B cell-containing samples are collected during immunization and analyzed by next-generation sequencing.
  3. Serum antibodies are affinity purified against the client-provided antigen and subjected to mass spectrometry analysis.
  4. Next-generation sequencing data and mass spectrometry data are analyzed by Alicanto discovery software to identify antigen-specific antibodies present in serum.

 

Deliverables:

  • At least 10 antibody sequences selected by Alicanto.
  • Full access to all antibody sequences detected.

 

Timeline - up to 20 weeks:

  • Immunization: 14 weeks
  • Data generation (NGS and LS-MS/MS): 4 weeks
  • Data analysis: 2 weeks

  • Rabbit monoclonal antibodies
  • Llama single chain antibodies
  • Alpaca single chain antibodies
  • Human monoclonal antibodies
