Desktop Genetics specialises in the design of custom high-quality libraries for CRISPR screening. We work with a network of expert partners to synthesise and assemble the highest quality reagents based on our designs. These partnerships allow us to tailor every library to your specific needs without incurring high customisation costs. With our pooled oligonucleotide CRISPR libraries, you have full control over the vector and viral production. We will design and synthesise the oligos to your specifications, using our machine learning-based design pipeline, and ship you the oligos for cloning into your vector of choice.

Using pooled CRISPR libraries, you can screen thousands of gene knock-outs in one experiment. Screening a pool of sgRNAs enables very high throughput functional genomics screening at a low cost. There is no need to screen thousands of constructs using expensive or complex liquid-handling robots.

Design:
Library designed using Desktop Genetics's machine-learning based sgRNA design algorithm.
sgRNAs are designed with minimal off-target effects and maximal activity.
Entire library is design by taking into consideration multiple published and un-published parameters.
sgRNAs can be designed to target protein functional domains, promoter regions, splice junctions, etc. - completely customised to your targets and cell line.

DNA:
Synthesized using Twist Bioscience's high-throughput oligonucleotide synthesis platform.
High quality oligo pool without randomizations and with minimal mutation rates (>0.2%).
Representation is maintained with massively parallel high throughput synthesis method to ensure similar quantities of each sgRNA oligonucleotide in the pool.

Vector:
Chose from an All-In-1 vector system or split Cas9/sgRNA 2-vector system.
Chose from constitutive or inducible (TET-ON3G) promoters for expression of Cas9 or sgRNA.
All vectors include dual selection markers (GFP & PuroR).
Pooled cloning is validated using NGS-assay to ensure maintained complexity and representation of sgRNA population.

Lentivirus:
Manufacture using Vectalys's high-quality production pipeline
Very high lentivirus allow for efficient delivery of sgRNA libraries into a wide range of cell types.
Tailor the purity and concentration of the lentivirus pools to your cell types (10E6 - 10E9 TU/mL)
Transduce primary cells with our VectaPremium Lentivirus.

Analysis:
Each sgRNA serves as an unambiguous sequenceable barcodes.
Screening assay is based on NGS sequencing of amplified gDNA from pool of cells.
Desktop Genetics can assist you in the analysis and deconvolution of your screening results.

Deliverables:
- design manifest file (CSV of all CRISPR sgRNAs and their scores, associated genes, and all properties);
- lentivirus pool, ready for transduction;
- NGS quality report on lentivirus library with representation and complexity analysis.
