This includes the selection of target sites in the gene based on our optimized algorithm that maximizes on-target nuclease activity and minimizes off-target activity, the design of nuclease vectors and the donor template. CRISPR/Cas9 injection into eggs

- Nuclease injection to obtain founders: The nuclease mRNA and donor vector will be co-injected into fertilized eggs, followed by implantation of the eggs into surrogate mothers to obtain offspring. In cases where the nuclease expression vectors are designed and constructed by Cyagen, we will inject as many eggs and/or target as many sites as needed to fulfill the guarantee. In cases where the nuclease expression vectors (or their mRNA products) are provided by the customer, we will inject a minimum of 200/300 eggs (based on strain) and screen pups for founders carrying desired mutation. If no founders are identified, more injections can be performed at an additional charge.

Founder screening
- Pups will be screened by PCR to identify knockin/floxed founders. Specifically, the site targeted by CRISPR will be PCR-amplified to reveal any insertions. Occasionally, an animal may be found to have both alleles of the target site mutated. For conditional knockout projects, mice carrying two loxP sites on the same allele flanking critical exon(s) are considered floxed.

Transgenic founder mice/rats breeding to F
- Cyagen will breed founder animals to wt animals of matching background, and screen F1 animals with desired mutation(s). A minimum of 2 F1 animals per line will be generated and delivered.
- Large cohorts of F1 or F2 animals can be generated upon request

ES cell gene targeted heterozygous mice breeding to homozygous mice
Standard KO/KI mice breeding service
- Cyagen will set up heterozygote x heterozygote breeding and screen pups by PCR for targeted mutation(s). Theoretically this breeding will produce 50% heterozygotes, 25% homozygotes and 25% wt animals. We will provide 2~4 homozygous mice per line. If no homozygotes are identified from a total of 30 pups screened, it is possible that the homozygous mutation may lead to embryonic lethality. If this is the case we will stop breeding and discuss the project with clients.

Conditional KO/KI mice breeding service
- Cyagen can inter-cross flox/wt mice to generate flox/flox mice, and can further breed them with Cre deleter of your choice to produce tissue-specific KO/KO (or KI/KI) mice. We will deliver 2~4 flox/flox mice or 2~4 tissue-specific KO/KO (or KI/KI) mice.

CRISPR-mediated genome editing mice/rats breeding
- Although animals generated by CRISPR-mediated genome editing carry targeted mutation(s), the founder animals are different from animals generated by ES cell gene targeting approaches in that most of the founder animals display some degree of mosaicism. Cyagen offers founder animal breeding service which allows us to deliver germline transmission-confirmed heterozygous/homozygous animals to researchers.
- We guarantee generation and delivery of a minimum of 3 heterozygotes. We can further inter-cross heterozygotes and deliver homozygous animals (a minimum of 2 per line). - Integration occurs as a single copy at each integration site. By contrast, in conventional transgenics, there are typically multiple copies integrated in tandem at each integration site. Scope of Work Performed

Knockout strategy design

  • Tell us the name of gene you wish to knockout, and we will design a nuclease-mediated strategy that meets your research needs. This includes the selection of target sites in the gene based on our optimized algorithm that maximizes on-target nuclease activity and minimizes off-target reactivity, and the design of nuclease expressing vector(s). For each gene, we will design vectors against at least two target sites in the gene to ensure success. Genotyping assays based on PCR and sequencing will also be designed for the screening of founders.

Nuclease expression vector construction

  • DNA vectors that express the desired nucleases will be constructed. Where needed, the efficacy of these vectors will be tested in cell culture.

Nuclease injection into eggs

  • Preparation of nuclease mRNA: Nuclease expression vectors will be transcribed in vitro. The resulting mRNA will be artificially capped and polyadenylated to facilitate its proper translation into protein in mammalian cells.
  • Nuclease injection to obtain founders: The nuclease mRNA(s) will be injected into fertilized eggs, followed by implantation of the eggs into surrogate mothers to obtain offspring. In cases where the nuclease expression vectors are designed and constructed by Cyagen, we will inject as many eggs and/or target as many sites as needed to fulfill the guarantee. In cases where the nuclease expression vectors (or their mRNA products) are provided by the customer, we will inject a minimum of 200/300 eggs (based on strain) and screen pups for founders carrying desired mutation. If no founders are identified, more injections can be performed at an additional charge.

Founder screening

  • Pups will be screened by PCR and sequencing to identify founders. Specifically, the site targeted by the nucleases will be PCR-amplified, followed by sequencing of the PCR product to reveal any mutations that might have occurred. Animals carrying frameshift deletions/insertions or critical exon(s) deletion on at least one allele are considered knockout founders. Occasionally, an animal can have both alleles of the target site mutated.

Breeding founders to obtain F1

  • For some projects, the generation of founder knockout animals is the end point. However, some customers wish to have us breed the founders further to obtain F1 animals. We will breed up to 3 founders to wildtype animals of matching strain background, and genotype their offspring to obtain F1 animals bearing the knockout allele.
