The Epigenetics Team at ChemPartner is led by senior scientists with broad assay development experience. To date, we have nearly 100 epigenetic enzyme assays, which are developed and optimized in house to provide our partners with fast delivery, flexible target selection and quality data. We have at our disposal diverse technology platforms including TR-FRET, AlphaLISA, Caliper mobility shift and radiometric assay for compound profiling, selectivity panels, mechanism of action studies and HTS. The targets encompass all classes including HKMT, HRMT, HKDM, HDAC, HAT, Bromodomain, Methyl reader and DNMT, among others. We continuously expand our offering in this exciting area.

ChemPartner is well-equipped with a variety of cell-based assays including Histone H3 Methyltransferase, which enable us to examine epigenetic target engagement and its functional consequences in cells. ChemPartner continually expands the scope of its technologies including BacMan/GFP/TR-FRET so that it is at the forefront of Epigenetics research.

In addition to the in vitroassays, we have also established over 100 xenograft models, many of which can be used in epigenetics studies. For instance, the SETDB1, CARM1 can be applied in breast cancer, and Ezh2, LSD1 for prostate cancer. These models have been used in compound testing for efficacy studies and PK/PD correlation.

SH (fluorescence in situ hybridization) is a cytogenetic technique to detect and localize the presence or absence of specific nucleotide sequences on chromosomes or RNA targets (mRNA, lncRNA and miRNA) in cells, circulating cells, and tissue samples. It can help define the spatial-temporal patterns of gene expression within cells and tissues. Recently, FISH has been more and more widely used to detect chromosomal alterations that underlie the development and progression of cancer, and pinpoints targets of new anti-cancer drugs.

ChemPartner offer high-quality and efficient service for FISH assay development, staining , imaging and data analysis/interpretation. So far, ChemPartner has handled hundreds of clinical patient tumor samples for FISH study and accumulated extensive hands-on working experience with FISH troubleshooting.

The readout wil be tumor volume, body weight, clinical signs, tumor wieght and tumor pictures at the study termination.

The Tumor Immunology Group at ChemPartner combines the expertise from Oncology and Immunology Groups and is led by leaders with extensive experience in Cancer Immunotherapy Drug Discovery.

More than 20 syngeneic tumor models in immune competent mice have been developed, including some of the most widely used ones, such as CT26, 4T1, B16F10, RENCA, EMT6, LLC and etc. They are used to study efficacy, mode of action (MOA), and tumor-host interaction. In addition to serum cytokine measurements, lymphocyte composition of CD4, CD8, Treg, and myeloid-derived suppressor cells from lymph tissues such as spleen, tumor draining lymph nodes (TDLNs), and tumor infiltrating lymphocytes (TILs) are analyzed using flow cytometry or immunohistochemistry (IHC) for MOA studies and biomarker discovery.

Primary immune assays using isolated T cells or other immune subsets have been developed to characterize or select lead antibodies, or to analyze mouse surrogate antibodies. T cell proliferation assays, intracellular cytokine staining of T cells by flow cytometry, or cytokine measurements from cultured T cell supernatant by ELISA serve as readouts from pharmacologically-treated naïve CD4 cells, total CD4 cells, enriched T cells, and mouse total splenocytes. Antigen-recall assay in PBMC is also available for analyzing T cell checkpoint immune modulators.

Our chemistry team has >10 years of experience in providing organic synthesis service, we are able to synthesize any organic molecule per our clients’ requests, in an efficient, convergent manner.

With >1000 experienced and skilled chemists, we make, QC, process and ship compounds from milligram to kilogram level, including reference compounds, building blocks and scaffolds, intermediates, final single compounds, antibody drug conjugates, and more.

For one-off synthesis work, we can provide economical and high quality custom synthesis as a fee-for-service (FFS) arrangement. For an on-going project, such as medicinal chemistry support, usage of a full-time-equivalent (FTE) arrangement provides dedicated scientists. Generally, the FTE service is more efficient for our clients than FFS service for repeated work.

Our library synthesis group uses parallel synthesizers, microwave reactors and prep scale HPLC to rapidly deliver high quality compounds for our clients projects.

We can design small property or target class focused libraries or larger DOS type libraries. We work closely with our computational and structural groups on design. Our designs our guided by structure based drug design when crystal structures are available, and our computational group for property based metrics. We can deliver compounds in a variety of formats or deliver them ready for screening to our in vitro biology group or in vitro DMPK group for assay.
