The Chemical Biology platform at the Biotechnology Centre Oslo (University of Oslo) has been in operation since 2008 and offers many unique services related to intracellular flow cytometry (particularly Phosphoflow) and protein-protein interaction (AlphaScreen). We have available more than 60,000 compounds, dispensed using the Echo 550, as well as having multiple robotic automation and assay systems.

We offer a fully-automated 384-well service for Phosphoflow studies. This allows quick turnaround times and increased reproducibility. Some example studies are described below.
HTS-Phosphoflow Screening - We have developed a fully automated 384-well system for processing samples for use in phosphoflow studies. By exploiting a close proximity to an active basic research group in immunology (28 publications utilising phosphoflow since 2008), the system has been developed over the past two years and extensively tested leading to true HTS-screening (16 level barcoding) and fast turn-around times for processing patient samples. Our large panel of phospho-antibodies are in constant use and samples are run on a LSR-Fortessa (BD) equipped with the 384-well loader.
Phosphoflow phosphorylation Monitoring - We can monitor baseline phosphorylation for patients involved in clinical research projects or clinical trial studies in order to better understand individual patient’s responses.
We have the BD LSR-Fortessa available with 384-well loader for use together with our automated Phosphoflow platform.
We offer 384-well HTS AlphaScreen technology to be used for disrupting protein-protein interactions (PPIs). The assay uses recombinant, tagged protein together with commercially available alphascreen beads. Compounds are available for use here or compounds can be provided by the customer.
Using the 384-well FLIPR from molecular devices, we can offer screening of targets that modify intracellular calcium levels such as ion channels and GPCRs. Cells are washed and loaded with calcium sensitive dye (Fluo-4) using the EL-406 plate washer from Biotek and data is analysed automatically using an excel macro. Cell lines are normally provided by the user. Discounts available for additional plates.
