Our sgRNA design includes parameters that optimize for on-target knockout while minimizing off-target cross reactivity. We can also include a panel of non-targeting, intron-targeting, and lethal sgRNA controls to provide reference standards when analyzing screening results.

Additionally, Cellecta can clone the designed sgRNA sequences into nearly any lentiviral vector.

You give us your list of targets, and we do the rest.

These lentiviral preparations can be directly added to cells in culture or directly in laboratory animals. The lentiviral particles fuse with the cell membranes and the lentiviral RNA enters the cell, is reverse-transcribed, and integrated into the cell’s DNA.

Lentiviral expression constructs packaged into pseudoviral particles can be transduced into cells with very high efficiency, approaching 100% in some cell types. Packaged lentiviral constructs can be transduced into even the most difficult to transfect cells, such as primary, stem, and differentiated cells with high efficiency.
We can provide various amounts of virus depending on your needs.

Viral titers are based on actual transduction of viral preparation into 293K cells. We do not use estimates based on PCR amplification from viral RNA or p21 detection of viral proteins, except with custom constructs that do not contain a fluorescent marker or antibiotic selection.

Our service is set up for packaging of our extensive line of lentiviral vectors. However, we can also package third-party vectors provided they are compatible with our packaging system.

Turnaround time is 2 weeks, and 25 ug plasmid DNA is provided. Constructs can be additionally packaged starting at $350 for 1 x 10^6 transduction units (TU or ifu) for single-vector sgRNA constructs containing Cas9 and $350 for 1 x 10^7 TU for two-vector sgRNA-only constructs. Turnaround time for packaged constructs is 2-3 weeks. Larger quantities may take longer to deliver.

You can also provide us with the CRISPR sequences you want, and we will make the insert, clone it, and sequence it to verify correct construction. Pricing is the same as above.

Single-Vector CRISPR/Cas9 System

The Single-Vector CRISPR/Cas9 system can be used for knockout of individual genes in vivo or in vitro by expressing both sgRNA (single guide RNA) and Cas9 nuclease from the same lentiviral vector.

  • Co-transduction not necessary since sgRNA and Cas9 nuclease are expressed in one vector
  • Lentiviral vectors integrate into the host cell genome and are passed onto daughter cells
  • Cells can be treated, grown for several passages, frozen, and thawed
  • The lentiviral construct remains in the host cells
  • Vector contains antibiotic selection to ensure stability

Two-vector CRISPR/Cas9 System

The Two-vector CRISPR/Cas9 system is typically used to perform functional knockout screens in vivo or in vitro by expressing sgRNA (single guide RNA) and Cas9 nuclease from different lentiviral vectors.

  • Pre-transduce with Cas9 lentivirus to decrease noise in your knockout screen
  • Make high-titer sgRNA lentivirus for efficient transduction

Pooled lentiviral-based libraries allow you to assay the effects of many thousands of effector constructs simultaneously in one experiment. Producing a reliable and effective screening tool with such complexity requires considerable expertise. Cellecta has extensively optimized library construction protocols and overcome a number of technical challenges to produce quality effector libraries. Over the last ten years, we have built and tested over 500 shRNA, CRISPR, barcode, and other effector libraries.

Built from Quality Components

  • Synthesized using Agilent's microarray-based oligonucleotide synthesis platform
  • Minimal mutation rate (typically ~0.2%, >95% correct shRNA active region)
  • Each oligo sequence in pool is fully defined without randomizations
  • Solid support synthesis minimizes bias by providing similar yields of each individual oligonucleotide

By taking advantage of Agilent's array-based oligonucleotide synthesis, we can readily make precisely defined large custom pooled libraries expressing many thousands of elements. Within approximately 2-3 months we can produce a completely new, high quality custom library.

The library vector can also be easily be customized, and the same library oligonucleotides can be cloned into more than one vector backbone (e.g., with an inducible and constitutive shRNA promoter).

Cellecta can help validate target genes by knocking down expression in your cells of interest. Cellecta provides complete services for validation of target genes including qRT-PCR, proliferation assays, reporter assays, and custom stable knockdown cell line creation. We design and make several constructs to the target gene, then test these to find the one with the best knockdown:

  • You tell us the RefSeq number or gene ID of the target gene you want to knockdown. We do the rest.
  • Best knockdown is measured by RT-PCR, and is usually better than 70%
  • We can provide you with the confirmed shRNA construct for the gene target
  • Alternatively, we can send you the stable pool we used for testing

Proliferation, Survival, or Viability Assays

  • Quantify growth rates, survival, or cell death by FACS analysis
  • Uses tet-inducible shRNA expression to compare growth rate of induced vs. uninduced cells

Turnaround time is 2 weeks, and 25 ug plasmid DNA is provided. Constructs can be additionally packaged starting at $350 for 1 x 10^7 transduction units (TU or ifu). Turnaround time for packaged constructs is an additional 2-3 weeks. Larger quantities may take longer to deliver.

  • Choose constitutive or inducible version of H1 or U6 shRNA promoters
  • Select GFP, RFP, PuroR, BleoR, NeoR, or Hygro-HK markers
  • Obtain constructs as plasmid or packaged lentiviral particles
  • Customize the cloning vector for an additional fee

To make an shRNA construct, we only need to know the RefSeq number. We can design any number of shRNA constructs that target the transcript. Our proprietary shRNA design delivers approximately 70% shRNAs that generate >70% transcript knockdown in standard cell lines as measured by qRT-PCR.

Alternatively, you can provide us with the shRNA sequence you want or reference an shRNA in one of our libraries, and we will make the insert, clone it, and sequence it to verify correct construction. Pricing is the same as above.

For validated shRNA expression constructs, we can design and make a validated shRNA expression construct that is confirmed by qRT-PCR to knock down any desired target gene transcript by at least 70% in the cell line of your choice. For this service, we design and make several constructs expressing different shRNAs to the target gene, test knockdown by qRT-PCR, and provide the most effective construct.
