Oncology Animal Models

The causes and treatments of cancer remain one of the biggest and most demanding areas of clinical treatment in the world. Due to our extensive background in cancer research, Buffalo BioLabs specializes in tumor model development and its applications. Located adjacent to the nation's very first cancer facility, Roswell Park Comprehensive Cancer Center, we are able to work directly with their renowned researchers and clinicians to provide insight on the clinical impact of our clients drug development programs. We offer orthotopic, subcutaneous, syngeneic, and xenograft models. Survival, cage-side observations, and tumor measurements can be used to evaluate disease progress in any model although some require tumor imaging. Other endpoints such as histopathology and biomarker screening can be combined with tumor models to gain additional insight regarding the efficacy of anticancer therapies. Discovery Pathology

A comprehensive assessment of pathological changes is integral to preclinical studies for determining safety, efficacy, and mechanism of action of novel drug candidates. Necropsy is performed to record and preserve gross morphological changes at the conclusion of in-life. Tissue and organs collected for analysis are preserved by snap freezing, formalin fixation, perfusion fixation, or fixed-frozen methods. Paraffin or cryo sample preparation is available for H&E and custom IHC staining. Services are performed by experienced histological technicians carefully adhering to all SOPs and study protocols and ensuring all samples are properly labeled, catalogued, and stored for in-house evaluation or shipment directly to Sponsor. Qualified histologist and pathologists are available to interpret staining results.

In vivo PK/PD Studies

Performing PK Studies are key to the selection of lead compounds with desirable bioavailability profiles for further investigation in drug development programs. Well-designed PK studies provide ADME (absorption, distribution, metabolism and elimination) data to assist in the study design. Cytotoxicity Assays

Buffalo BioLabs offers cytotoxicity assays in 96 well format for a broad range of cancer cell lines using methylene blue and resazurin based readout. Serial dilutions of up to 9 doses are used in duplicate or triplicate, depending on the sensitivity of the methodology. IC50s are calculated using GraphPad Prism software on a minimum of 3 independent experiments. Alternative assays include clonogenic assays on a minimum of 5 drug concentrations and growth in soft agar. Assays can be adapted to client’s cell line(s) of choice.

Acute systemic toxicity includes a single exposure with a 72-hour observation period and is most commonly performed. Repeat or continuous exposure, classified as subacute, chronic, & subchronic  are also available.
