AxoSim fabricates 3D hydrogel scaffolds which encourage the development of a mixed population of cells, introducing polarity, migration, axon outgrowth and myelination. These conditions replicate the environment found in living organisms. Our models can be cultured for months, and have the potential to be tailored based on different therapeutic applications.
AxoSim's facilities are equipped to measure changes in electrophysiology, neurite outgrowth, nerve density, fiber type, degree of myelination, and cell integrity following either acute or chronic exposure to compounds of interest. Range finding experiments can also be performed for concentration dependent effects.
During the process, AxoSim manages all aspects of the research, culminating with an intensive high-level report and presentation discussing all available data. During the client presentation, potential implications will be discussed and any client questions will be addressed.
In addition to our 3D assays, AxoSim provides both high content imaging and 2D electrophysiological assays. We can run high-throughput image based analysis to determine cell viability, neurite outgrowth, proliferation, and glial interactions. For comparison to functional characteristics, we can also run high-throughput multielectrode array (MEA) assays to determine changes in firing rate, bursting characteristics, and response to stimuli.
We use standard fluorescent and confocal microscopy to measure in vitro changes in 3D neurite outgrowth after exposure of our Nerve-on-a-Chip(TM) platform to neurotoxic, neuroprotective, or neuroregenerative compounds. Population-level electrophysiology, as measured using the compound action potential, is performed on our Nerve-on-a-Chip(TM) platform. Changes in peak amplitude, conduction velocity, latency, and integral after drug administration correlate to clinically relevant mechanisms of action for neurotoxicity, neuroprotection, and neuroregeneration.
