Modern molecular and cellular biology has resulted in the ability to express proteins of interest in a variety of host cell lines. Aragen’s proven success in stable protein expression – even for the most difficult to express proteins – is based on decades of experience and success with hundreds of variant molecules.

Experience, Experience, Experience

Aragen’s founding scientist began his career expressing proteins in the early 1980’s as part of a team at Genentech. Since then, he and the team at Aragen have gained experience with hundreds of diverse molecules and continue to optimize the expression vectors, workflows and processes around stable cell line development and protein expression. This has resulted in the development of a robust, in house, CHO-DG44 platform for secreted proteins and other molecules of interest.

Diverse Products

The ability to stably express proteins and produce the desired amount of product depends on a large number of factors. Depending on the project, only a few hundred micrograms of protein may be needed while other projects may require the manufacture of kilogram quantities of protein. Difficult to Express Proteins

Successfully expressing and purifying proteins can be difficult. Given the breadth and depth of our cell line development and protein expression experience, it is not surprising that the Aragen team is often called upon to stably express and purify novel molecules that are challenging to express either due to folding problems, toxicity to hosts, purification issues or any of a wide range of other problems. Aragen’s approach of continually optimizing and improving processes and assays enables our scientists to custom tailor the cell line development process to your specific requirements.

Regulatory Friendly and Royalty Free

Aragen’s CHO-DG44 process has complete freedom to operate and comes with no clinical milestones or commercial royalties. In addition, the core technology has been reviewed many times over many years by the various regulatory agencies of the world. As such, this platform is well-documented and comes with very little regulatory risk.

What to Expect – Timelines and Titers

For protein expression projects requiring large amount of protein, the volumetric titer is critical to meeting the budget for manufacturing efforts. Aragen’s platform is very competitive with respect to its timeline to delivery and the titers obtained.

In Vivo Efficacy Services

At Aragen Bioscience, we offer specialized in vivo services to support your pre-clinical research needs. Our campus includes an 8,500 square foot dedicated animal facility that is equipped to provide a variety of animal studies and ex vivo analysis services. Unlike other CROs that specialize in only one or two focus areas, we have invested the time and resources to establish a wide variety of working animal models that support multiple therapeutic/disease areas including oncology, infectious disease, fibrosis, respiratory disease and immunology/inflammation. We also undertake non-GLP pharmacokinetic, pharmacodynamics and toxicity rodent studies.

The initial screening of hybridoma’s enables the identification of panels of monoclonal antibodies of interest. The furhter triage these panels to identify those exhibiting more specific characteristics, such as binding affinity, species and/or family member cross reactivity, cell based activity and the like, requires the significantly more protein product that afforded by the small scale cultures available during the initial screening stages.

Aragen Biosciences team has expertise in various methods for the scale up of antibody production. For example, the production of the monocolonal antibodies of interest can be performed either by the scale up of the hybridoma’s themselves or via the cloning and expression of the monoclonal antibody using recombinant techniques. If a recombinant method is chosen, one might also elect to isotype switch the antibody or perhaps generate a human-mouse chimeric molecule should a human Fc domain be required for further efficacy evaluations. Importantly, Aragen Bioscience can produce and purify monoclonal antibodies from the micro gram to gram scale.

As is well known, the characterization of monoclonal antibodies and their function is a very complex task. There are various biophysical and molecular techniques (e.g. Affinity to target, stability, aggregation propensity, etc.) as well as efficacy measurements (e.g. ADCC, Growth Inhibition, in vivo models) that may be required. Aragen, as an end-to-end provider of antibody discovery services, enable it’s clients and partners the ability to perform such complex studies within a single contract research organization (CRO).

Many hybridoma cell lines, especially those identified early on in a discovery program, can be very unstable. This phenotype can be due to many factors including clonality, chromosomal rearrangement and other metabolic stresses.

In order to minimize the impact this may have, the Antibody Discovery team at Aragen Bioscience strongly believes that hybridoma culture showing any signs of instability (e.g. early hybridoma cell lines or older lines losing expression) should be subcloned immediately. Our robust hybridoma subcloning process has also rescued a few high value hyridomas for clients.

In addition to in-house-developed hybridomas, we also offer subcloning for hybridomas supplied by our clients. Similar to the antibody fusion screening, our subclone screening includes high-throughput ELISA and FACS based assays. Samples can also be shipped to clients for additional testing.

Upon completion, at least 3-5 subcloned hybridomas will banked (cryopreserved) and shipped to the client.

It has been said that “you get what you screen for”. Hence, we believe that the screening cascade must be carefully designed and executed in order to identify the rare antibody events that are inherent to antibody discovery programs.

Hybridoma screening is one of the most important parts of the antibody development work flow. Aragen antibody discovery scientists work collaboratively with the client to design the most appropriate screening strategy to identify target specific and high affinity antibodies. Our extensive screening procedures cover:

  • Sero-conversion
  • Antigen reactivity
  • Epitope binning
  • Affinity determination using the ForteBio OctetRED system
  • Cell based binding and activity
  • Cross reactivity

In addition to our standard fusions using whole cells from spleen and lymph nodes, we also offer fusions using isolated B-cells to enrich the antibody secreting B-cell population. Although hybridoma culture supernatants are primarily screened in-house using our high-throughput ELISA and FACS-based assays, we also encourage clients to perform additional screening at their facility to confirm specificity and also to confirm the presence of the protein. Proper controls are included in the screening assays to verify specificity and cross-reactivity.

The importance of hybridoma technology primarily lies in its ability to harness the host’s innate ability to generate diverse and high affinity antibodies when challenged with an antigen. However, the nature of that challenge can significantly alter the outcome. Aragen offers a wide variety of immunization schemes using a diverse set of immunogens including peptides, whole cells, purified recombinant proteins, and cDNA (plasmids).

We understand that manipulation of the immune response is critical in generating the right antibody. Therefore, we offer several immunization strategies including:

  • Standard bi-weekly immunizations
  • RIMMS (Rapid Immunization at Multiple Sites) method
  • Decoy immunizations
  • cDNA immunizations

Customized Immunization Regimens to Meet Your Needs

All immunizations are performed at our on-site Animal Facility that provides instant access to different types of hosts including mice, rats and hamsters, including Armenian hamsters. We also use different strains of mice for higher epitope diversity. Our Animal Facility staff has extensive experience in performing subcutaneous, intravenous, intramuscular and intraperitoneal injections using a variety of adjuvants including CFA, IFA, CpG-based adjuvants, RIBI and TiterMax. In addition to in-house testing of anti-sera, samples will also be shipped to the clients for additional confirmation of specific immune response.

Antigen design and synthesis is a critical initial step for a successful antibody discovery project. The specific application of the antibody to be developed plays a key role in determining the immunogen design strategy. Given Aragen’s many years of experience, we have successfully executed on a very diverse set of antigen types including:

  • Peptides
  • Recombinant proteins
  • Whole cells
  • cDNA (plasmids)

Our Protein Science group has extensive experience in recombinant protein synthesis using bacterial and mammalian expression systems. We routinely generate a variety of recombinant proteins including Fc-fusion proteins, epitope-tagged fusion proteins (6X-HIS, GST, FLAG, HA etc.), un-tagged proteins, KLH- or BSA-conjugated proteins, full length or truncated proteins and protein domains. We also work with a third party supplier to generate synthetic peptides including KLH-, BSA- and OVA-conjugated peptides.

Our Cell Line Development group has experience generating transient and stable cell lines which can also be used as immunogens primarily for cell surface antigens. We also offer the design and construction of plasmids for cDNA immunizations.

Extensive quality control measures are executed while generating the antigens. We understand that some investigators prefer generating their own antigens. Therefore, to meet our clients’ needs, we also accept antigens synthesized off-site.

The ability to purify proteins to a high level of purity is critical in order to investigate their physical and biological properties. Some proteins are relatively easy to purify using robust, well established affinity chromatography methods. These include antibodies and Fc fusion molecules as well as proteins containing a recombinant purification element such as the 6X His tag. However, many molecules are not afforded such elements and more complex purification strategies must be developed. This is often the case with many novel fusion proteins as well as other molecules such as enzymes and cytokines.

Aragen Bioscience has experience in the routine purification of secreted monoclonal antibodies, intracellular tagged proteins, novel cytokine fusion molecules and many other molecules produced by a variety of expression platforms:

Whether your project starts from an idea, electronic sequences or a standard reagent, Aragen scientists will first determine the appropriate cloning and expression system for your protein of interest. For truly novel proteins, the Protein Science team will guide your project design based on both their structural and practical (e.g. purification) experience. Our team will also recommend additional process development activities that will help to increase production titer, alter product quality, and/or improve purification efficiencies. In addition to novel or difficult to express proteins, Aragen also has a proven track record for purification of hundreds of antibodies, tagged molecules and more traditional proteins.

As the demand for protein product can very significantly, our cell culture capabilities range from a few milliliters up to one hundred liters. These volumes are used to express from microgram to milligram or gram levels (25 – 100 grams). Our AKTA based downstream purification infrastructure is highly scaleable to flexibly and reproducibly handle diverse projects.

The technique provides a valuable approach for harnessing the diversity of the host immune system to discover novel antibodies with unique properties.

Our team of Antibody Discovery scientists have extensive experience in developing hybridoma-based mAbs to a wide range of therapeutic and diagnostic targets. We have successfully developed antibodies to a variety of targets including cytoplasmic, secreted and membrane bound proteins as well as anti-idiotype antibodies. Aragen’s uniqueness lies in our ability to carry out antibody development projects from idea to pre-clinical stage. We work very closely with the clients on complete project design, offering several options for successful antibody development.

Antibody Development Workflow

Attention to detail is critical at every step of the antibody development and discovery process. If the antigen is not designed properly, the animals may never seroconvert or, in some cases, may only seroconvert to an immuno-dominant epitope reducing the diversity of the antibody repertoire that is assayed. Our team has experience with a broad range of antigens and immunization schemes that are critical for a target-specific response. By leveraging our core technology platforms, we are able to execute the appropriate assays dictated by the screen which are typically a combination of ELISA and cell-based methods.

Antibodies for Various Applications
