Antibody Core Facility

The primary objective of our facility is to produce monoclonal antibody-producing hybridoma cell lines for UNC investigators. The core develops monoclonal antibodies in mice, rats and hamsters. The core can assist in the screening of hybridomas of interest by ELISA or flow cytometry. The facility can also produce large amounts of crude antibody in vitro for investigators as well as purify both polyclonal and monoclonal antibodies from tissue culture media or ascites.

Antibody Purification

We typically utilize the advanced capabilities of our Protein Expression and Purification facility with protein A, G or A/G affinity chromatography to purify your antibody. Post purification we can exchange the antibody into a buffer you choose and concentrate it to your specifications.

The Antibody Core has developed a method to generate antibody from a monoclonal hybridoma cell line using an in vitro method that compares favorably to the ascities method. The Antibody Core utilizes several different methodologies (suspension culture, cell line flasks, nunc cell factories) and specially formulated medias for antibody production. Typically, antibody concentrations are about 0.2 mg/mL using this technique with variations between 0.05 - 2mg/mL. These concentrations are about 5 to 10 fold what is commonly obtained using other methods. There is a lot of variation of antibody yield from a production due to hybridoma differences such as lines that are inherently low producers, lines that adapt poorly to the production media, cell growth characteristics, etc.

For very poor producing hybridomas there is an alternative to increase your antibody yields. Working with our Protein Expression and Purification facility we can attempt to isolate the DNA coding sequences for the antibodies and develop a recombinant mammalian expression system. Please contact the director for more information.

For most hybridomas, it will take about 2 weeks to adapt the cell line to the media and 3 weeks to produce the high-yield final material. If requested, the Antibody Core can purify the antibodies from the raw material.
