iPS Cell Generation Services

ALSTEM works as an extension of a customer’s team to provide iPS cell generation services that can be quickly and seamlessly implemented.
ALSTEM's stem cell scientists are experts in stem cell biology, including reprogramming of somatic cells into induced pluripotent stem cells (iPSCs). ALSTEM’s iPS cell generation services include expert scientific consultation to plan your reprogramming project, fast turn-around with scheduled milestone status updates to inform you of the progress of your cells, and comprehensive advice in maintaining your reprogrammed iPS cells upon delivery to your laboratory.

Multiple reprogramming technology options
Both Viral and non-viral reprogramming systems are available which includes episomal DNA reprogramming and retrovirus reprogramming. These technologies have been adopted and developed in close collaboration with leading iPS cell pioneers. Our extensive range of products allows for customization of individual transcription factors, cell culture conditions, and delivery systems for the optimization of reprogramming parameters.

Service Details

A. Retrovirus-based iPSC reprogramming protocol
- Retroviral reprogramming using standard Oct4, Sox2, c-Myc, and Klf4 Factors
- iPSC colonies are selected based on morphology
- AP Staining and immunostaining of ESC markers (Nanog or Oct4, SSEA3 and TRA-1-81 included)
- 5 vials iPSC colonies will be provided (>2 x 10^5 cells/vial)
- Timelines for human iPSCs: 3-4 months and mouse iPSCs: 2-3 months

B. Episomal vector-based iPSC reprogramming protocol
- Episomal DNA reprogramming using Oct4, Sox2, c-Myc, and Klf4 factors with small molecules
- iPSC colonies are selected based on morphology
- AP Staining and immunostaining of ESC markers (Nanog or Oct4, SSEA3 and TRA-1-81 included)
- 5 vials iPSC colonies will be provided (>2 x 10^5 cells/vial)
- Timelines for human iPSCs: 4-5 months

Required Materials
Customer-desired cells, 1 vial of typical storage size, and indication of appropriate growth medium and conditions. Currently most iPSC lines were created from fibroblast-type of primary cells; other cell types may not be appropriate using the current protocols.

Deliverables
We typically aim to deliver 5 vials of iPSC colonies (>2 x 10^5 cells/vial). Customer specified quantity of cells or number of lines can be discussed.

Turnaround Time
3 - 5 months generally, actual time will vary based on method chosen and additional customer requirements.
