Nanobody Libraries

Single Domain Antibody Library

VHH can be raised from an immune library or a naïve library. 

Advantages of Naïve Library:

  •    No immunogenicity issues: applicable for toxic, lethal or transmissible antigen proteins.
  •    Short turnaround time: only 3 - 4 weeks to get positive antibody sequences.
  •    Animal protection: no animals required.
  •    High diversity libraries: we provide various naïve library with high diversity.

Disadvantages:

  •    Lower affinity and specificity

Our naïve library was constructed from PBMCs isolated from healthy, non-immunized alpacas. Antibody VHH-genes were amplified from mRNA and cloned into a phage display vector. The library consists of 108 individual transformants with more than 90 % unique sequences. We leverage this established library along with our advanced screening platform to extend the epitope coverage and help clients find the optimal monoclonal antibody candidates.

Phage Display Immune Libraries

In terms of the advanced phage display technology, AlpaLife has capabilities for the construction of VHH or VNAR based single domain antibody libraries through immunization of camel, alpaca or shark. In comparison with naïve libraries, immune libraries usually produce antibodies of greater affinity, thus avoiding time-consuming in vitro antibody affinity maturation effort. Antibodies from naïve libraries frequently require affinity maturation before they are useful due to affinity issues. Furthermore, in comparison with naïve libraries, which usually require a size of 1-10 billion independent clones to be useful, an immune library can be much smaller. Experiences showed that immune libraries with a size of 1-10 million variants could be sufficient to produce excellent antibodies.

AlpaLife offers customized library construction services under M13 filamentous phage display system. Through DNA manipulation, numerous gene variants can be created and constructed as phage display library. Our scientists are confident in generating high-quality libraries with a diversity of 10^8.

AlpaLife nanobody production service includes analysis of customer-provided antigen, antigen modification, immunization of one animal, library construction and screening, delivery of Bacterial Glycerol Stock of positive clones and antibodies sequences.

Workflow:

Phase I. Animal immunization. The phase includes antigen modification for immunization, immunization of one animal (alpaca camel or shark) with client-provided antigen, blood collection (20 ml), and ELISA analysis, PBMC isolation and preservation. Turnaround period: 8-12 weeks.

Phase II. Phage Display Library Construction. Antibody cDNA generation via RNA isolation, PCR amplification, and diverse antibody library construction. Turnaround period: 3 weeks.

Phase III. Library Screening. Immune library Phage display screening, ELISA verification, identification of positive clones, antibody sequences. Turnaround period: 6 weeks.

To meet the growing industry demands for nano-antibody development and animal immunization, our company established professional and specialized platforms for alpaca breeding and immunization to minimize the cost and risk for VHH and VNAR antibodies global developers.

AlpaLife has built 9 animal breeding and researching centers across China. With high standard facilities and professional animal caring staff, those centers provide a large number of healthy alpacas, sharks and camels with clear immune-background for VHH antibody and VNAR antibody development. Our company's animal centers are open and welcome our customers come by themselves to visit, sample and inspect animals.

Give us your antigen proteins and we will do all the rest for you.

In general workflow, we perform 4-6 times of immunization. After the last immunization, our staff will collect 20 mL blood samples from the immunized animal and extract peripheral blood mononuclear cells.

Standard workflow for alpaca immunization:

0 ~2nd-day. Analyzing antigen protein and finishing protein analysis report.

2nd ~4th-day. Collecting the blood sample before immunization.

5th-day. Antigen preparation with complete Freund's adjuvant and injection.

20th-day. Antigen preparation with incomplete Freund's adjuvant and injection.

26th-day. Collecting blood samples and extracting peripheral blood lymphocytes.

35th-day. Antigen preparation with incomplete Freund's adjuvant and injection.

42nd-day. Collecting blood samples and separating serum.

50th-day. Antigen preparation with incomplete Freund's adjuvant and injection.

56th-day. Collecting 20 mL blood samples and extracting peripheral blood mononuclear cells (PBMC).

60th-day. Analyzing serum by ELISA. Providing project reports.

We can customize immunization periods and frequency according to customer requirements.

Requirements for antigen proteins:

Purity: High purity of antigen proteins is critical for animal’s immune response, we suggest to immunize protein with purity > 90% or peptide with purity > 95%, length > 12 amino acids.

Concentration: >1 mg/mL without toxic components like azide.

Amount: 4 mg of soluble protein or 10 mg of peptide usually is enough, low immunogenicity antigen could need more than that.

Shipping: packaging with ice dry when shipping.
